Background and objectivesBreast cancer is one of the malignant tumors which have a high prevalence and mortality rate in males through out the world, among which its prevalence and mortality rate in China have increased in resent years. The therapies for beast cancers include mastectomy,radiotherapy,chemotherapy,hormonal and biology therapy. But recurrence and metastasis of the tumor still affect the patient's lifetime. Therefore, our tentative study on the pathologic mechanism of breast cancer and its therapies are necessary. Survivin is a novel member of apoptosis protein family. Its structure and character are very different from other members of inhibitor of apoptosis proteins(IAP). Survivin is expressed in the G2/M phase of the cell cycle in a cycle-regulated manner. Survivin binds the terminal effecter proteases, caspase-3 and caspase-7 to inhibit cell apoptosis. Formation of three-dimensional vascular tubes in vitro is associated with strong induction of Survivin in endothelial cells. The building of the blood vessel plays a very important role in the growth and metastasis of the tumor. The endothelial cell-specific mitogen vascular endothelial growth factor(VEGF) has been found to be expressed by tumor cells through autocrine and paracrine pathwaies. It is the most important advanced vascular factor. Oncogene promotion and tumor suppression gene mutation can induce tumorgenesis. Tumor suppression gene P53 plays a very important role in dignosis and prognosis of breast cancer. The aim of this study is to evaluate the relationship between the expression of Survivin, VEGF and P53 and clinicopathologocal factors in breast cancer, which will provide theoretical basis for the evaluation, therapy and prognosis of breast cancer.MethodsThe expression of Survivin, VEGF and p53 was evaluated in 60 cases of breast cancer ,20 cases of hyperplasia of mammary gland and 10 cases paracacinoma normal tissues by immunohistochemical technique(SP).The correlation among the expression of Survivin,VEGF and P53 and the relation with the clinical characteristics of breast cancer were analyzed. The data were analyzed by software SPSS 16.0, x~2-test was used to analyze the difference between different groups. P value <0.05 was considered as statistically significant.Results1. The positive rates of Survivin were 76.7%,18.2%,0%in breast cancer, hyperplasia of mammary gland and paracacinoma normal tissues, respectively(P<0.05).The expression of Survivin was significantly correlated with the histological grade of breast cancer and Her2 in breast cancer.2. The positive rates of VEGF were 73.3%,31.8%,10%in breast cancer, hyperplasia of mammary gland and paracacinoma normal tissues, respectively(P<0.05).The expression of VEGF was significantly correlated with lymphonodes mestastasis, the histological grade of breast cancer, Her2 and clinical staging in breast cancer.3. The positive rates of P53 were 51.7 % ,13.6 % ,0% in breast cancer, hyperplasia of mammary gland and paracacinoma normal tissues,respectively(P<0.05).The expression of P53 was significantly correlated with tumor size, lymphonodes mestastasis, the histological grade of breast cancer , Her2 and clinical staging in breast cancer.4. The expression of Survivin was closely correlated with the expression of VEGF and P53. in breast cancerConclusions1. The expression of Survivin in breast cancer shows a significant rise and a stong relationship with the histological grade and Her2 ,which suggests that the high expression of Survivin may play an key role in growth and development of breast cancer2. The high expression of VEGF in breast cancer shows a close correlation with lymphonodes mestastasis, the histological grade of breast cancer, Her2 and clinical staging ,which suggests that VEGF expression is closely related to growth,metastasis,and development of breast cancer.3. The high expression of P53 in breast cancer shows a close correlation with tumor size, lymphonodes mestastasis, the histological grade of breast cancer , Her2 and clinical staging , which suggests that P53 expression may play an important role in dignosis and prognosis of breast cancer.4. There exists a positive correlation of the positive expression between VEGF and Survivin, which suggests VEGF may be an important factor in inducing the up-regulation of survivin.It also may favor angiogenesis of breast cancer and make it easier to invade and metastasize.5. There exists a positive correlation of the positive expression between P53 and Survivin, which suggests P53 may promote the expression of Survivin. Survivin and P53 may co-operate cell apoptosis in breast cancer.6. There is a close correlation of Survivin expression,VEGF and P53 in breast cancer, which suggests Survivin, an important factor in cell anti-apoptosis and angiogenesis, may become a new target for dignosis, prognosis and gene therapy of breast cancer.
