Objective:To establish the method of isolation and cultivation of rat bone marrow mesenchymal stem cells(BMSCs) in vitro.To evaluate the simultaneity expression efficacy of rat bone marrow stromal cells(BMSCs) transfected with bone morphogenetic protein 2(BMP2) and vascular endothelial growth factor(VEGF) genes.Methods:BMSCs from rats were isolated and cultured in vitro by using the whole bone marrow adherence method.The BMSCs were transfected with BMP2 and VEGF165 by liposome-mediated method.The co-espression of BMP2 and VEGF165 genes were determined by Enzyme-linked immuno sorbent assay (Elise) and immunohistochemical analysis.Results:The cells transfected with BMP2 and VEGF165 indeed could produce the proteins of BMP2 and VEGF165.Conclusion:the BMSCs transfected with BMP2 and VEGF165 genes will be expression simultaneously of rat bone marrow mesenchymal stem cells.
