Objective: Metastasis is the leading cause of death in clinic.Metastasis is a complex process unique to cancer.Tumor cells shed from the tumor block, adhesion and penetration of vascular, reenter the blood circulation, after penetrating the vessel wall into the normal organization cells in growth and reproduction and final form of metastasis. This process requires the participation of a variety of molecules. The sugar molecules on tumor cell surface is related to tumor adhesion and metastasis.Fucosyltra- nsferases is a kind of catalytic enzymes to fucose synthesis of oligosaccharides,The major role ofα1, 3-fucosyltransferase (Fut4) is the specific synthase with LewisY (LeY) in cell surface .Lewis antigens is a carbohydrate antigens found by glycolipid and glycoprotein chain on the cell surface. Fut4 joins in the fucose reaction of Lewis carbohydrate synthesis in the last step. The major role of which is to transfer Fuc from GDP-Fuc to N-acetyl glucosamine of N-acetyl lactose unit in sugar chain. Fut4 is expression in the white blood cells and epithelial cells,the abnormal expression of Fut4 in tumor cells, such as in pancreatic cancer and colorectal cancer ,the expression of Fut4 is significantly high. squamous cell carcinoma causes canceration by the squamous epithelial cells. Clinically, such as skin cancer, laryngeal cancer, lung cancer, cervical cancer, breast cancer.In China, there is a high incidence of squamous cell carcinoma, therefore, revealed the development of squamous cell carcinoma, research in both sugar chains of cell surface and the relationship between cell transfer and adhesion about scc12 squamous cell, provides a theoretical basis to prevention and treatment of squamous cell carcinoma.This study by observing increase the expression of Fut4 affect behavior of malignant transformation about scc12 squamous cell .Discussion on mechanism of Fut4 promote tumor metastasis. Methods: The experiment choose scc12 squamous cancer cells ,extract Fut4-cDNA and empty vector,transfer them to scc12 cell separately by cell transfection experiments. Detected mRNA and protein expression of Fut4 after transfection by RT-PCR and Western blot. Observed them about migration and invasion by cell scratch test separately. Analyze migration and adhesion of scc12 cell by cell adhesion test. So compared the scc12 cell of transfected Fut4-cDNA to the cell of untransfected plasmid.Results: The results of RT-PCR and Western blot demonstrate that mRNA and protein expression of Fut4 in scc12 cells by transfected Fut4-cDNA were higher than in scc12 cell of untransfected plasmid, The results of cell scratch test and cell adhesion test show that migration and invasion about the cell of transfected Fut4-cDNA were higher than the cell of untransfected plasmid.Conclusion:The expression of Fut4 play important role in migration course of the scc12 cells.The expression of Fut4 increased,lead to metastasis and adhesion of scc12 increased, which benefit for finding out the role about fucosyltransferase in the behavior of squamous cell carcinoma malignant transformation.It will provide new ideas and means to metastasis and adhesion of tumor by using glycobiology.
