Background: Adipose tissue transplantation has became an vital method in soft tissue reconstruction with the development of plastic surgery. However, the lower survival rate after the adipose tissue transplantation limited its clinical application. The lidocaine and adrenaline are used in tumescent anesthesia during the adipose tissue obtaining. Therefore,there are a lot of lidocaine and epinephrine in the adipose tissue. Many experiments has approved that the lidociane(0.08%) and adrenaline (0.0002%) depress the adipocite activity.There was no relevent report confirm whether the depressed effect could be stronged.Objective: To evaluate the depressed effect to adipocyte by lidociane(0.08%) and adrenaline (0.0002%) by detecting the creatine kinase level change peripheral adipocyte and adipociyte survival rate by trypan blue staining under microscope.To provide the evidence how to deal with the factor during adipose tissue transplantation. Content:The adipose tissue was obtained from the patient in general hospital in shen yang military region. The adipocyte was divided into four groups: divided into A (saline group), B (0.08% lidocaine), C (0.0002 % epinephrine group), D (0.08% lidocaine and 0.0002%epinephrine group)in 12 patients, respectively. Then the lidociane(0.08%) and adrenaline (0.0002%) was detected in 1 hour, 2 hours, 4 hours. The results was dealed with SPSS13 . 0.1.Creatine kinase (CK) detection. Creatine kinase (CK) is the key kinase to energy metabolism in adipose tissue embolism. Creatine kinase (CK) measurement of peripheral fat granule cells can reflect the adipocyte changes by injury, also reflect the activity of adipose tissue.2.the percentage of trypan blue staining of adipose cells are observated under the inverted microscope .trypan blue staining principle is that when the fetal cells be injury or death, trypan blue dye can penetrate the fetal adipose cells membrane degeneration, and that is binding and coloring the disintegration of the DNA .however the living cells can prevent the child trypan blue dye into the cell, so the dead cells were stained light blue, while the living cells resist infection. By observing the percentage of adipose cells trypan blue staining of fetal survival, to evaluate the treatment factors on the activity of fat cells.Results:1.Creatine kinase levels of change: At the same time point, the CK value changes were different significantly between the groups and the control group and different between each group.( P≤0.01 in Table II): CK value changes significantly between group D than the others.2.Tire change in the percentage of fat cells with rypan blue staining observed. under Inverted microscope.At 1 hour, 2 hours, 4 hours time point, the percentage change were compared between each group and the control group. There were no significant difference between each group(P> 0.05).Conclusion:1. 0.08% lidocaine solution, 0.0002% epinephrine in tumescent anesthesia in clinic has depressed effect on adipocyte activity. The effect with use of both of them was significantly higher than use of each of them. The effect with use of 0.08% lidocaine on the fat cells was higher significantly compared with 0.0002% epinephrine.2.When umescent anesthesia solution with 0.0002% and 0.08% lidocaine and epinephrine on fat cells lasting 4 h has no impact on depress fat cell survival.3. The fat particles obtained need to be rinsed to remove the swelling solution of lidocaine and epinephrine to obtain satisfactory results.
