| Water-soluble bee pollen polysaccharides from Rosa rugosa was reported inthis paper. All fractions were analyzed by gel-permeation, monosaccharidecomposition, physico-chemical property and molecular weight distribution.The water-soluble polysaccharides from the bee pollen of Rosa rugosa wereextracted with hot water, precipitated by 80% ethanol, and a crude polysaccharidefraction, referred as WRPP (for Water-soluble bee Pollen Polysaccharides of Rosarugosa ), was obtained with a yield of 7.8% (w/w). WRPP contained 52.1% of totalsugar, 9.7% uronic acid and less than 6% protein. Sugar composition analysis byHPLC indicated that WRPP consisted of Man:Rha:GlcA:GalA:Glc:Gal:Ara=2.6:3.4:1:12.1:11.6:21.4:47.9. WRPP was loaded on DEAE-Cellulose column andobtained one neutral fraction (WRPP-N) and two acidic fractions (WRPP -1 andWRPP -2). All fractions were analyzed by the contents of sugar, contents of uronicacid, monosaccharide composition and molecular weight distribution.Analytical results showed that WRPP-N was a mixture of glucans andarabinogalactans. It was consisted of Glc, Man, Gal and Ara as a ratio of 34.3:25:36.7:4.WRPP-1 was mainly composed of Rha(3%),GalA(12.4%),Gal (24.7%) and Ara(53.9%). The amount of Gal and Ara acount for over 70% of total sugars.The ratio ofRha/GalA was 1:4 and Gal/Ara were 1:2. The results indicated that type-Irhamnogalacturonan (RG-I) and homogalacturonan (HG) domain were existed asmain-chain, and arabinogalactan as side-chains.WRPP-2 was mainly composed of Rha, GalA, Gal and Ara as a ratio of7.8:23:15:48.7, while the Man, Glc and GlcA were as a little proportion. The resultssuggested that RG-I and HG domains were existed as main chain, arabinogalactan andarabinan as side chains. Gel permeation chromatography and high permeationchromatography showed the molecular weights approximately ranging from 5.0×10~5to 2.5×10~4 Da. It has more RG-I and HG domains than WRPP-1. |
