Objective:Breast cancer is one of the most common malignant tumors among females. In our country, the trend of the incidence seems to increase every year. Therefore, people are paying more and more attention to how to give better treatments to the patients thereby improving the cure rate and survival rate. Among all kind of cancers, one type, known as highly malignant, TNBC (triple negative breast carcinoma),lacks the conventional treatment such as Endocrine therapy and Specific therapy, for its ER, PR and Her-z are all negative expressions. So, the treatment of TNBC turns out to be the hotspot and difficulty. Through the research on the expressions of BRCA-1 and PARP-1 in the tissue, TNBC, non-TNBC, breast fibriadenoma and breast normal tissues, and through the study on the relationship between the two genes and the relevant clinical feature, we explore its function in prophase diagnosis, treatment and the evaluation of prognosis, in order to improve the cure rate and survival rate of patients with TNBC.Methods:1. Collect the tumors and the normal tissues beside the tumors in surgeries in the People's Hospital of Tangshan City. There are 21 cases of TNBC, 36 cases of non-TNBC, 32 cases of normal tissues and 32 cases of breast fibriadenoma. No treatments are given to these cases before the collection. All the tissues are put into fridge in the temperature of -80℃in half an hour.2. Distill the general RNA in the breast tissue in the method of Trizol. Analyze the integrality, density and purity. Proved to be eligible in the test, the RNA is reversed transcription. Lastly, apply the method of real-time quantitative Polymerase Chain Reaction PCR to test the expressions of mRNA with BRCA-1 and PARP-1 in TNBC, non-TNBC, tissues between the tumors, breast fibriadenoma and normal breast tissue. Take GAPDH as the reference gene. Analyze the data by means of 2-△△C t.Results:1. For BRCA-1, the relative expression value of mRNA in cancer tissue and breast cancer adjacent tissue of TNBC patient are (0.734±0.226,2.372±0.781), which shows there is a significant difference(P<0.05); and the value in non-TNBC patient are (1.037±0.659,2.711±1.924), which also shows there is a significant difference(P<0.05); meanwhile, the relative expression value of mRNA in breast fibriadenoma and breast normal tissue are (1.463±0.337,3.389±2.090), which shows there is significant difference(P<0.01).2. For PARP-1, the relative expression value of mRNA in cancer tissue and breast cancer adjacent tissue of TNBC patient are (0.810±0.172,0.162±0.088), which shows there is a significant difference(P<0.05); and the value in non-TNBC patient are (0.694±0.047,0.140±0.111), which also shows there is a significant difference(P<0.05); meanwhile, the relative expression value of mRNA in breast fibriadenoma and breast normal tissue are (0.273±0.026,0.101±0.017), which shows there is a significant difference(P<0.05).3. In breast cancer, the group of the relative expression value of BRCA-1 which is higher than average value, the relative expression value of PARP-1 is lower than the other, which shows there is a significant difference (r=-0.575,P<0.05),and both have Anti-related.4. The low expression of BRCA-1 relates to patients are attacked low ages, high histological grade and positive lymph node metestasis, and it also relates to the negative expression of molecular markers ER/PR high expression.5. The low expression of PARP-1 relates to high histological grade and positive lymph node metestasis, and it also relates to the negative expression of molecular markers ER/PR and p53 high expression. Conclusions:1. As the development and deteriorate of breast disease, the expression of BRCA-1 has the decrease tendency, which indicate high relationship between BRCA-1 and breast cancer, especially TNBC, and the BRCA-1 may has an effect on the treatment of TNBC.2. PAEP-1 may has an effect during the growing, deterioration and transforming of the breast cancer, the high expression of TNBC indicate that the application of PARP-1 in breast cancer treatment may get a better curative effect.3. The high expression of PARP-1 relates to the low expression of BRCA-1 in TNBC. And the application of PARP-1 inhibitor in clinical may get a better curative effect on BRCA-1 low expression breast cancer.4.The expression of mRNA shows an increase tendency of BRCA-1 in TNBC cancer tissue, non-TNBC cancer tissue, breast fibriadenoma, and breast normal tissue; on the other hand, the expression of mRNA of...
