Objective:To study the expressions and significance of Ki67,BRCA-1 in triple negative breast cancer(TNBC) and analyze the relationship between Ki67 and BRCA-1 expression in TNBC, according to investigate clinical pathology features of TNBC such as age, tumor size, etc. At the same time, to research the difference of Ki67 and BRCA-1 expressed in Luminal B Her(+), Her-2 overexpression and TNBC. Methods:322 breast cancer cases admitted to Fujian Provincial Hospital were collected by from January 2012 to December 2016, of which 78 cases were discovered as TNBC. None of them had done preoperative treatment such as chemotherapy, radiation therapy, targeted therapy and endocrine therapy. The expressions of Ki67, BRCA-1 in the breast cancer were detected by Elivsion two-step immunohistochemistry method. And the relation of Ki67, BRCA-1 with clinical pathology features were analyzed thought SPSS.Results: 1. There were 19 Luminal A(5.7%), 143 Luminal B Her-2(-)(43.1%), 38 Luminal B Her-2(+)(11.4%), 54 Her-2 over-expression(16.3%) and 78 TNBC(23.5%). 2. The differences of KI67 and BRCA-1 in Luminal B Her-2(+), Her-2 overexpression and TNBC are significant.(P<0.05). 3. 78 TNBC data sets, there was correlation(P<0.05) between Ki67 expression, tumor size, lymph node metastasis, the number of lymph node metastasis, histological grading and TNM stage. The Ki67 expression of TNBC with lymph node metastasis(84.2%) was much higher than non-metastasis group(51.2%). Meanwhile, Ki67 expression in Histological grade III tumors(86.6%) was higher than histological grade I-II(53.8%). The Ki67 expression in TNM III tumors(91.4%) was higher than TNM I(78.8%) and TNM II(56.3%). There was no significant correlation(P>0.05) between Ki67 expression, with age of onset and menstruation. 4. In 78 TNBC data sets, there was correlation(P<0.05) between BRCA-1 expression and molecular classifications, such as lymph node metastasis, the number of lymph node metastasis, histological grading and TNM stage. The BRCA-1 positive expression with lymph node metastasis(75.4%) was much higher than non-metastasis group(47.6%). Meanwhile, BRCA-1 positive expression of histological grade III(73.1%) was higher than grade I-II(38.5%). The BRCA-1 positive expression of TNM III(61.5%) is higher than that of TNM II(42.6%). There was no significant correlation(P>0.05) between Ki67 expression, with age of onset, tumor size and menstruation. 5. The expression of CK5/6 was correlated with Ki67 expression(P<0.05). However, it was not related to BRCA-1(P>0.05). 6. As there is enhancement of Ki67 expression, the positive expression of BRCA-1 increases(P<0.01, r=0.456). Conclusion: 1. There are expression differences of Ki67 and BRCA-1 in Luminal B Her(+), Her-2 overexpression and TNBC are statistically significant. The expressional rates of Ki67 and BRCA-1 in TNBC are higher than Luminal B Her(+) and Her-2 over expression subtype. 2. Lymph node metastasis, the number of lymph node metastasis, histologic grade and TNM stage are correlated with the expression of Ki67 and BRCA-1 in TNBC. As histologic grade and TNM stage increase, the expression and the number of lymph node metastasis rise. Hereby, the expression of Ki67 and BRCA-1 may be used to evaluate of prognosis and guidance of cancer treatment. 3. The expression of CK5/6 is correlating with Ki67 expression, but not related to BRCA-1 positive expression. In fact, there is lack of relevant literature reports as reference to make a further discussion and develop on this topic.
