Objective:To study the effect and role of leptin on myocardial ischemia - reperfusion injury (MIRI) in mouse.Method:Twenty four male rat were randomized into 4 group:①the sham-operation group;②the myocardial ischemia reperfusion group(MIRI group),established by blocking left coronary artery anterior descending branch for 30 min followed by reperfusion③the leptin pretreatment group(Leptin+MIRI group)established by injecting intraperitoneally leptin at 30 min before ischemia;④the leptin post-treatment group(MIRI +Leptin group)established by injecting intraperitoneally leptin at 30 min after reperfusion.Blood and heart samples were harvested from each mouse at 120 min after reperfusion.We then analyzed the samples for leption level associated with MIRI.Result:the leptin post-treatment group(MIRI +Leptin group) myocardial infarct size〔(39.43±2.06)%〕,myocardial leptin levels〔(23.1±2.95)μg/g〕and serum leptin levels〔(38.37±1.98)μg/L〕significantly higher than the myocardial ischemia reperfusion group(MIRI group)myocardial infarct size〔(33.20±2.12)%〕,myocardial leptin levels〔(15.50±3.14)μg/g〕and serum leptin levels〔( 29.49±2.01μg/L〕; the leptin pretreatment group(Leptin+MIRI group) myocardial infarct size〔(9.20±1.85)%〕,myocardial leptin levels〔(8.26±3.29)μg/g〕and serum leptin levels〔(10.31±1.88)μg/L〕significantly lower than the myocardial ischemia reperfusion group(MIRI group)myocardial infarct size〔(33.20±2.12)%〕,myocardial leptin levels〔(15.50±3.14)μg/g〕and serum leptin levels〔(29.49±2.01μg/L〕; the leptin post-treatment group(MIRI+Leptin group) lactate dehydrogenase (LDH)〔(1067.4±123.2)U/L〕,aspartate aminotransferase (AST)〔(402.9±55.0)U/L〕,Troponin T(TnT)〔(0.98±0.13)μg/g〕levels significantly higher than the myocardial ischemia reperfusion group (MIRI group) actate dehydrogenase (LDH)〔( 884.5±104.3 ) U/L〕,aspartate aminotransferase (AST〔)(327.1±44.9)U/L〕,Troponin T(TnT)〔(0.72±0.10)μg/g〕; the leptin pretreatment group(Leptin+MIRI group) creatine kinase (CK)〔(3231.7±640.1)U/L〕,lactate dehydrogenase (LDH)〔(610.4±86.0)U/L〕,aspartate aminotransferase (AST)〔(202.3±29.1)U/L〕,Troponin T(TnT)〔(0.51±0.09)μg/g〕levels significantly lower than the myocardial ischemia reperfusion group (MIRI group)creatine kinase (CK)〔(5310.3±849.4)U/L〕,actate dehydrogenase (LDH)〔(884.5±104.3)U/L〕,aspartate aminotransferase (AST)〔(327.1±44.9)U/L〕,Troponin T(TnT)〔(0.72±0.10)μg/g〕。Conclusion:Treatment and high level of leptin increased the myocardial ischemia / reperfusion injury, leptin pretreatment reduced myocardial ischemia -reperfusion injury。...
