Objective: The paper evaluates the influences from long-term blood donation behaviors on humoral immunity through quantitative analyses on blood group anti-body and study on immunity state of organism including CD4, CD8, CD34 and NK cells.Methods: 38 blood donators (whole blood or PCs) with annual average blood donation frequency≥2 are chosen in the experiment, non-AB blood group male donators with accumulated frequency≥30 are chosen as observation group, 34 non-AB blood group males without blood donation experiences are chosen as contrast group, for questionnaire investigations and blood test on mental status,psychological state,social dysfunction and living attitude etc. The blood test including the use of FCM for quantitative anti-body analysis of IgG anti-A(B) in serum and CD4, CD8, CD34 and NK cell test。At the same time,repeatability and recovery test method was used to test the method of the use of FCM for quantitative anti-body analysis of IgG anti-A(B) in serum . Sysmex fully automatic blood cell counter for counting of blood cell, red blood cell, white blood cell and hemoglobin. SPSS12.0 statistical software is used for experimental data analysis; a separate sample t test is used in testing and comparing the differences between the various observation indices of the observation group and contrast group.Results: As for the health condition, anxiety and insomnia, social dysfunction and depression degree, etc.There are not evident differences between the long-term blood donation group and the non-blood donation group (P>0.05). There are evident differences on IgG anti-body between B group, the average value of the contrast group is lower than the observation group. There are not evident differences between A group and O group.The average value of the relative standard deviation of the batch is 2.97, the regression confficient( R2 )inrecovery test is above 0.95. There are not evident differences between the two groups on the percentage of CD4/CD8 and CD34+ (P>0.05). However, the NK cell number of observation group is evidently higher than the contrast group, but the average value of the platelet count is lower than the contrast group (still within the normal range), and the differences between the two groups are evident (P<0.05).Conclusion:The method of assay the anti-IgG contents using flow cytometry is effective and reliable. Long-terms blood donation can influence the natural antibody generation statistically compare to the control. Long-terms blood donation may promote the generation of NK. Long-terms blood donation may reduce the number of platelets in peripheral blood.
