OBJECTIVE : A novel cross-linked Cyclosporine A nanoparticles blend membrane based on silk fibroin and polyvinyl alcohol monomer were prepared, for the treatment of Psoriasis.METHODS:(1) CyA-NPs were prepared by emulsion-solvent diffusion technique and freeze-dried. Properties of CyA nanopartles such as morphological form, diameter, distridution, drug loading, encaosulation efficiency were investigated; (2) An optimal prescription of the membrane was obtained with different monomer ratios, plasticizers concentration; (3) Prepare the membrane of CyA-NP with different types and concentration of enhancers individually or blend together, the cumulative permeation quantity (Q) and steady-state penetrating rate (JS) and relative parameters were evaluated for screening the optimizing prescripeion of membrance; (4) The optimized membrane was characterized by SEM, FTIR, X-ray and the mechanical properties of membrane such as the tensile strength, elongation percentage at break and weight loss ratio were determined; (5) The percutaneous permeation profiles of CyA-NPs from membranes were studied in vivo in rats; (6) The biocompatibility and toxicity of prepared membrane were investigate by animal experiment.RESULTS:(1) The nanoparticles produced displayed smooth surface, consistern diameters,mean diameter was 43.9±0.82 nm, the encapsulation efficiency reached to 99%; (2) When silk fibroin and polyvinyl alcohol in a ratio of 9:1 and plasticizers concentration in 2%, the cross-linked membrane showed good physical property and good film-forming property, flexibility, homogeneity; (3) The most appropriate enhancers added into membrane were 1% azone and 2% propylene glycol united in this research. And the steady-state penetrating rate was 6.59μg·cm-2·h-1. The enhancement ratio was 2.59 fold compare with control group,cumulative permeation quantity in 24h was 2.81 times compared with the membrane without penetration enhancer; (4) The cross-linked membrane had favourable mocrostructure and nanoparticles were uniformly distributed over internal and surface of membrane; (5) The percutaneous permeation kinetic models of CyA nanopaticles membranes were studied in vivo, the results demonstrate that these CyA nanopaticles membranes significantly improved the percutaneous permeation of CyA; (6) The membranes was an safety drug dilivery system.CONCLUSION:Silk fibroin protein was a good biomaterial for drug delivery. The silk fibroin-polyvinyl alcohol cross-linked membrane exhibits superior mechanical properties and film-forming property. The CyA-NPs cross-linked membranes significantly improved the percutaneous permeation of CyA.
