Objective To explore the therapeutic effect of BMSCs combined with BDNF on chronic spinal cord injuries of adult rats.Methods 48 adult rats were subjected to a weight driven implant injury and randomly divided into four groups. After four weeks of SCI, each group was injected respectively with BMSCs+BDNF, BMSCs, BDNF and DMEM(the control group). After 1,2,4 weeks of transplantation, BBB scores was used to measure the motive function of SCI rats.4 weeks after transplantation, immunohistochemical technique was used to measure the expression of growth-associated protein-43 and the distribution of BMSCs.Results After 4 weeks of transplantation, BMSCs were found surviving in the injured spinal cord of BMSCs+BDNF group and BMSCs group. Comparing with other groups, the morphological repairing of BMSCs+BDNF group was much better and the positive expression area was much larger (F=35.57, q=4.97~25.84, p<0.05). BBB scores showed that functional recovery of BMSCs+BDNF group was best in the four groups (F=27.51, q=3.89~11.57, p<0.05), and BDNF could promote the therapeutic effect of BMSCs(F=13.67, P<0.01).Conclusion It is more effective for structural and functional recovery of chronic spinal cord injury adopting therapy of BMSCs+BDNF and it was proved that BDNF could improve the repairing of BMSCs.
