Camphor is a large evergreen tree Lauraceae, alias camphor, camphor wood, black camphor, linalyl, Fan Zhang, Hong Rui, camphor son, lobular camphor, its leaves contain a large number of lignanoids compounds. Lignans is a kind of natural compounds by two molecules of phenylpropanoid derivatives (C6-C3monomer) polymerization. It has many biological activities as antioxidant, anti-tumor, regulate plasma cholesterol, protect the liver, anti-inflammatory, antiviral, antibacterial. It has great significance in prevent and treat the cancer, cardiovascular disease, aging and other disease.This paper studies the camphor leaves lignan extraction process, the antioxidant activity and anti-tumor activity of the crude product that extract from the lignans, the contents include:(1) The process conditions of the optimized ultrasound-assisted extraction of lignans from camphor leaves. Camphor leaves as raw material, 95% ethanol solution as extraction solvent, use the ultrasound-assisted extraction method to extract lignans in the camphor leaves. Respectively discuss the effect of liquid feed ratio, ultrasonic power, ultrasonic time, extraction temperature , extraction time, these five factors on the yield of crude lignans. Basic on the single factor experiment choice the orthogonal design. Single factor experiments and orthogonal test results show that ultrasound- assisted extraction of lignans from camphor leaves optimum conditions: solid-liquid ratio of 1:25, the ultrasonic power is 300w, ultrasonic time of 30min, the extraction temperature is 50℃, extraction time of 75min.(2) Studied the camphor leaves lignan in vitro antioxidant. Using L-ascorbic acid and BHT as a control, use the potassium ferricyanide(K3[Fe(CN)6]) reduction method for the determination of camphor leaves lignans reduction ability; Fenton method for the determination of camphor leaves lignans remove hydroxyl radical (·OH) capabilities; Adjacent pyrogallol method to determine the ability of clearing the superoxide anion radical (O2-·) Spectrophotometry to determine the ability of clearing the DPPH free radicals (DPPH·) . Results in vitro antioxidant experimental showed that the camphor leaves lignan has good reduction ability,is good in clearing hydroxyl radicals (·OH), superoxide anion radical (O2-·), DPPH free radicals (DPPH·) Various abilities are higher than the general antioxidants,(3) Studied in camphor leaves lignan in vitro antitumor effect. Using MTT rapid colorimetric determination of camphor leaves lignans on human cervical cancer cells (Hela), human hepatoma cells (SMMC-7721) and human gastric cancer cells (SGC-7901) about the anti-tumor activity of these three kinds of cancer cells, and had a study on the camphor leaves lignan of anti-tumor mechanism of action. The results showed that lignans have inhibition on human cervical cancer cells (Hela), human hepatoma cells (SMMC-7721) and human gastric cancer cells (SGC-7901) these three kinds of cancer cells, showed strong inhibition on human cervical cancer cells, human hepatoma cells, the inhibitory effect on human gastric cancer cells is not obvious.
