Objective To investigate the effects of Panax ginseng on cell apoptosis and expression of SOD and MDA in human umbilical vein endothelial cells(HUVEC) exposed to intermittent high glucose and to search for the possible protective effect of Panax ginseng on HUVEC.Methods Take the human umbilical vein endothelial cells (HUVEC) in vitro as the research object to detect the vitality of cells in different concentrations of Panax ginseng by MTT. According to the requirements of the experiment, choose the best three concentrations of Panax ginseng and HUVEC which growing in the best period to go on the next step. Then divide the cells into three groups:the normal control group, the intermittent high glucose group (5.5mmol/L or 25mmol/L, high glucose 3h and low glucose 2h in turn by three times, and the high glucose treated all the night), the intermittent high glucose+Panax ginseng group in three different concentrations(12.5ug/ml,25ug/ml,50ug/ml). When the endothelial cells was incubated in different groups for 48h, the apoptosis rate of it was detected by Annexin V-FITC/PI, the activity of surperoxide dismutase (SOD) and the expression of malondialdehyde (MDA) in HUVEC was investigated by xanthine oxidase methods and thiobarbituric acid methods respectively.Results Compared with the normal control group, the rate of HUVEC apoptosis under the intermittent high glucose group was increased. And this group also has high content of MDA and the level of SOD in it was decreased obviously (P<0.001). Compare with the intermittent high glucose group, the low, medium and high concentration Panax ginseng groups had decreased apoptosis rate and MDA expression of HUVEC; the activity of SOD in these groups were all increased (P<0.05). But there were no significant differences between the normal group and the Panax ginseng groups (P>0.05). And there were not any significant differences during the three Panax ginseng groups in different concentrations (P>0.05), too. Conclusion Panax ginseng can restrain cell apoptosis and the secretion of MDA, promote the level of SOD in HUVEC. All above showed that Panax ginseng might be able to protect HUVEC from being injured under the intermittent high glucose. And it will be helpful to the treatment study of the diabetic cardiovascular complications.
