Effects The Traditional Chinese Medicine By Two Step Sequential Therapy On The Airway Trissue Of The Airway Inflammation And Remodeling Asthmatic Mice

Objective To establish the airway inflammation and remodeling asthmatic mice model, studied the effects of Ginseng-Schisandrae decoction and Xiaoqinglong decoction by two step sequential therapy on the airway inflammation and remodeling of the asthmatic mice, and to discuss its possible mechanism according comparison.Methods Eighty healthy female Kunming mice were divided into eight groups randomly (n=10, in each group):airway inflammation normal control group(Ⅰa), Airway inflammation model group(Ⅰb), Airway inflammation budesonide treatment group(Ⅰc), Airway inflammation two step sequential therapy group(Ⅰd), airway remodeling normal control group(Ⅱa), airway remodeling model group(Ⅱb), airway remodeling budesonide treatment group(Ⅱc), airway remodeling two step sequential therapy group(Ⅱd), The asthmatic model were established by ovalbumin, at the same time, budesonide Ginseng-Schisandrae decoction and Xiaoqinglong decoction were given to the treatment groups, Then the airway inflammation and remodeling in airway tissue was observed by PAS, HE staining, The level of nuclear factor-KB(NF-κB), GATA- 3,Matrix Metalloproteinases-9 (MMP-9)and Tissue Iihibitor Metalloproteinase-1(TIMP-1) in airway tissue were detected by immunohistochemical method.Results①HE staining:In groupⅠa:There was no or little inflammatory cells infiltration, and the bronchial wall was completing and smooth;GroupⅠb had a large number of inflammatory cells invasion such as eosinophils, neutrophils and lymphocytes around the bronchial wall, and airway epithelial cell were damaged, the pathological changes in groupsⅠc,Ⅰd significantly reduced compared with that of groupⅠb; In groupⅡa, there was no or little inflammatory cells infiltration, and the bronchial wall was completing and smooth; Inflammatory cells invasion such as eosinophils and lymphocytes can be seen around the bronchial wall in groupⅡb, there were also evidence of airway epithelium damaged, basement membrane and smooth muscle thickening, the pathological changes in groupsⅡc,Ⅱd significantly reduced compared with that of groupⅡb.②PAS staining:The score of PAS staining in groupⅡb was significantly increased compared with that of groupⅠb (P<0.01); There was no significant difference between groupⅡa and groupⅠa (P>0.05); The score of PAS staining in groupⅡc were significantly increased compared with that of groupⅠc (P <0.01); While that of was significantly increased in groupⅡd compared with that of groupⅠd (P<0.01);③NF-κB:The NF-κB of lung tissue were lower in groupⅡb than that of group I b (P<0.01); There was no significant difference between groupⅡa and groupⅠa(P>0.05); The NF-κB of lung tissue were lower in groupⅡc than that of in groupⅠc(P<0.01);While that of were lower in groupⅡd than that of groupⅠd (P<0.01).④GATA-3:The level of GATA-3 was decreased in groupⅡb compared with that of groupⅠb(P<0.01); There was no significant difference between groupⅡa and groupⅠa(P>0.05); The level of GATA-3 was significantly decreased in groupⅡc compared with that of groupⅠc(P<0.01); While that of was significantly decreased in groupⅡd compared with that of groupⅠd(P<0.01);⑤MMP-9:There was no significant difference between the acute and airway remodeling asthmatic mice (P>0.05).⑥TIMP-1: The TIMP-1 of lung tissue were higher in groupⅡb than that of groupⅠb(P<0.01); There was no significant difference between groupⅡa and groupⅠa(P>0.05); The TIMP-1 of lung tissue were higher in groupⅡc than that of groupⅠc (P<0.01);While that of groupⅡd were higher than that of groupⅠd(P<0.01).⑦the ratio of MMP-9/TIMP-1: The ratio of MMP-9/TIMP-1 was lower in the groupⅡb than that of groupⅠb (P<0.01), and there was no significant difference betweenⅡa andⅠa(P>0.05);The ratio of MMP-9/TIMP-1 was lower in the groupⅡc than that of groupⅠc(P<0.01); While that of groupⅡd was lower than that of groupⅠd(P<0.01).Conclusions①The study showed that Ginseng-Schisandrae decoction and Xiaoqinglong decoction by two step sequential therapy can significantly reduce the airway inflammation and airway remodeling of asthmatic mice in the cellular and molecular level Respectively;②Two step sequential therapy reduced airway inflammation and airway remodeling by NF-κB, GATA-3 and MMP-9/TIMP-1;③The study provided the basis for chinese medicine pharmacology in treating asthma.
