ObjectiveTo study the effect of bone mineral density (BMD) in lumbar spines, femoral neck, trochanter and proximal femur, biomechanics of lumbar spine and left proximal femur and plasma of all rats was collected to determine concentrations in estradiol and testosterone by radioimmunoassay of orchidectomized rats with zoledronic acid. And we contrast to the collators to understand the efforts of zoledronic acid on the less of bone. This may be able to afford a new way to cure OP by zoledronic acid.Material and Method1. Animals:We chose 36 male mouse of Wistar, SPF grade, weight rang from 230 gram to 250 gram, age from three to four months (provided by LU KANG Medical Corporation, Number of certificate of quality was SCXR 20080002). We randomized divide them to three groups Sham group with 12 mouse, orchidectomized (Orch) group with 12 mouse and orchidectomized+zoledronic acid (Orch+ZC 20μg/kg) group with 12 mouse by weight. We stared the study after two weeks adapting living and end in the eighteen week.Orch and Orch+ZC groups disinfected the skin of scrotum by iodine and alcohol under anaesthesia by 5 percents phenobarbital sodium. And then we carved the skin from the middle of the mediastina and cut the tunicae vaginalis. We separated the testicle and the epididymides and cut the testicle. Then we put the epididymides into the scrotum and oversew the incision. The Sham group was cut the tunicae vaginalis and separated the testicle and the epididymides (but not cut). Then we put it into the scrotum.2. Method of Medication:After the operation, we hyped ZC 20μg/kg into the mouse, mixed with 0.9% normal saline about 5ml. Sham and Orch group were hyped with the same volume.3. We dropped the mouse to the eighteen week and killed them kindly by drawing blood under 5% phenobarbital sodium (7.5mg/100g).We anatomized the animals and took the forth and fifth lumbar with no parenchyma. Using 0.9% normal saline cleaned the bone and using dual energy X—ray absorptiometry of Hologic QDR-2000+DEXA examined the bone density of L4-5,neck, troch and pro-femur. And then we used software to analysis. We did bomechanics of the forth lumbar and thighbone.4 The index and method:4.1 Weight:We took the weight every week and evaluated the factors.4.2 BMD:We used dual energy X—ray absorptiometry of Hologic QDR-2000+DEXA examined the bone density of L4-5,neck, troch, pro-femur and thighbone.4.3 Bomechanics:We used material testing machine of Instron-8874 to examined the bomechanics of the forth lumbar and thighbone.4.4 Statistics:We used software of SPSS 10.0 to analysis the data.Results18 weeks later, the weight of all the rats was increase. The Sham group was increased most with 112 gram and the Orch+ZC group was increased lest with 92 gram, with no difference, compared with the Sham and Orch group(P>0.05). The BMD in umbar spines, femoral neck, trochanter and proximal femur of the Orch group decreased, with statistic ally significant difference, compared with the Sham and Orch+ZC group (P<0.01). The BMD in the same parts of the Orch+ZC group was the highest among the groups, but not significantly different from that of the Sham group. The M-load, M-displacement, M-strain and M-stress of Orch group decreased, with statistic significant difference, compared with Sham and Orch+ZC group (P<0.01). But Orch+ZC group was not significantly different from that of Sham group. The M-stress, M-load decreased, with statistic significant difference, compared with the Sham and Orch+ZC group (P<0.01) The M-displacement, E-lastic decreased, with statistic difference, compared with Sham and Orch+ZC group (P<0.05). Orch+ZC group was not significantly different from that of the Sham group.ConclusionZoledronic acid can prevents osteoporosis in orchidectemized rats. And zoledronic acid can prevent osteoporosis in orchidectemized rats also.
