Objective To investigate the anti-tumor effect of Zhonggan Mixture and its mechanism.Methods Hepatoma H22 and S180 tumor-bearing ICR mice were established to evaluate the tumor inhibition effect of Zhonggan Mixture. Further,. ICR mice of S180 ascites carcinoma model was established to evaluate the life prolonging effect of Zhonggan Mixture. The expression of TGF-β,RⅡ, NF-κB, CyclinDl and VEGF mRNA were detected by quantitative real-time polymerase chain reaction (QRT-PCR) in tumor tissues.Results①Zhonggan Mixture significantly inhibited tumor growth in the mice bearing hepatoma H22 and S180, and the inhibitory rate of the low, mid and high-dose groups and Cyclophosphamide (CTX) group were 17.72%,33.99%,23.73%,43.95% and 29.59%,31.34%,1 1.17%,3.92% respectively. The mid-dose group displayed the most potent anti-tumor effect.②The life prolonging rate of the low, mid and high-dose groups and CTX group were 6.35%,16.95%,7.43%,17.46%.The mid-dose group displayed the most potent anti-tumor effect.③Zhonggan Mixture could up-regulate the expression levels of TGF-β1RⅡmRNA in tumor tissues from the mice bearing hepatoma H22. Compared with saline control group, the effects of the mid dose groups were the most prominent and showed significantly different in statistics(P<0.01).④Zhonggan Mixture could down-regulate the expression levels of NF-κB mRNA in tumor tissues from the mice bearing hepatoma H22. Compared with saline control group, the effects of the high dose groups were the most prominent and showed significantly different in statistics (p<0.01).⑤Zhonggan Mixture could down-regulate the expression levels of VEGF mRNA in tumor tissues from the mice bearing Hepatoma H22. Compared with saline control group, the effect of the mid and high-dose group was the most prominent and showed significantly different in statistics (P<0.01).⑥Zhonggan Mixture could down-regulate the expression levels of cyclinD1 mRNA in tumor tissues from the mice bearing hepatoma H22. Compared with saline control group, the effects of the high dose groups were the most prominent and showed significantly different in statistics (P<0.01).Conclunsion Our results indicate that Zhonggan Mixture has notable anti-tumor effects, and the possible mechanism of Zhonggan Mixture inhibiting proliferation and inducing apoptosis were associated with down-regulating the expression levels of NF-κB, CyclinD1 and VEGF m RNA and up-regulating the expression levels of TGF-β1RⅡmRNA in tumor tissues.
