Background material:early in the 256BC,there was a description about "royal surgeon" in the historical documents of Zhou dynasty,which we call cancer nowadays. With its incidence keep increasing, it is now one of the most malignant disease world wide. With the development of our study and knowledge, we know that it is the infinite proliferation of normal cells that makes the occurrence of malignant tumor, which is closely related with Environmental pollution, genetic defection and some other factors. With the improvement of our live standard,environmental governance and continuous development of antineoplastic therapy, though its incidence get some control, the effect is not optimistic. And there are many problems such as high toxicity, expensive price and drug resistance, which lead to the high expense low body quality of the patients,some even abandoning therapy. so minimizing the side-effect of chemotherapy drugs on the basis of a rather effect, and improving the immune function become the goal medical workers try hard to get. Only in this way they can help patient realize the "survival with tumor" living condition in a long term. Traditional Chinese medicine (TCM) is used more and more in the malignant tumor prevention and treatment both at home and aboard, because its high safety. low toxicity, more source of drugs, low price are its advantages. Because of its gratifying achievements in single anti-tumor drugs, prescriptions, compounds,it has become the one of the most important and effective means in comprehensive treatment of malignant tumors.Yiliuning preparation is a prescription which has a long-term experience in clinical practice of our department,consisting of Rubescens, Acanthopanax,Solanum nigrum and Duchesnea indica. It is a pure Chinese herbal mainly used in digestive system tumors, especially in esophageal cancinoma,gastrocardiac carcinoma,liver cancer,pancreatic cancer etc which makes no obvious side effects so far. It could obviously inhibit the growth of diversified tumor cells including esophageal cancer Eca-109 cells,gastric cancer SGC-7901 cells,pancreatic cancer BBxpc-3 cells,and liver cancer Bel-7402 cells,breast cancer MDA-MB-231 cells, lung cancer NCI-H520,lung cancer A549 cells, cervical cancer Hela cells. This experiment is on the basis of early studies on anti-tumor in vitro. we compared the Yiliuning with different sorts of chemotherapy drugs the dose-effect relationships and effects in vivo experiment,in order to provide experimental evidences for better clinical effect.Objective:Comparing Yiliunig preparation with other chemotherapy drugs in efficacy.Means:This experiment model is divided into five parts. we will select one sort of western type of chemotherapy drugs as positive control in each part.Adding NS to s180 animal tumor beads lets it become cell suspension for cell density of 10*107/ml, inoculating 0.2ml cell suspension in theright axillary subcutaneous of male mouse in the condition of no special pathogens in order to establish transplanted tumor model, the male mouse is a sort of kunming with general level,4 weeks old and their weight are 20 gram around. The above models will be randomly divided into eight groups. These groups are as follows:control group, high,medium, low dosage group of Yiliuning preparation, chemotherapy group and chemotherapeutic drugs being respectively combined with high,medium,low dosage group of Yiliuning preparation respectively. Concrete dosage will be interpreted in each stage.During this experiment,we will observe vital signs,general conditions,weight and changes in tumor weight and so on. After being inoculated, all groups will be given drugs for continuous ten days and tumor tissues of which will be cut in order to weigh and calculate inhibition ratio after the last 24 hours.Results:1,all groups are in good condition with none death during this experiment. Different sizes of miliary nodules can be seen in vaccination site gradually after 4 days for inoculating, diameter reaching about 0.5*0.5 cm which keep growing after 7 days for inoculating. The ratio of tumor formation is 100 percentage, so manufacturing tumor model is successful with a 100% ration of tumor formation.2,Positive control groups and groups with different dosages of Yiliuning preparation all can inhibit s180 transplanted tumor increasing in mice and the size and the weight of transplanted tumor in other 7 groups are lower than the control group. DDP is chosen as positive control:he tumor inhibition ratios of low, medium, high dosage groups of Yiliuning preparation are 34.36%,35.23%,39.05% respectively while the tumor inhibition ratios of DDP group,DDP combining with low, medium, high dosage group of Yiliuning preparation are 60.48%,60.48%, 67.62%,70.95%.We elect CTX as the representative of Alkylating agent. The tumor inhibition ratios of low, medium, high dosage group of Yiliuning preparation are 34.16% 35.19% 38.85% respectively while the tumor inhibition ratios of DDP group,DDP combining with low, medium, high dosage group of Yiliuning preparation are 54.17%,64.29%,74.29%,79.05%. We choose Xeloda as the representative of anti-metabolic drugs. the tumor inhibition ratios of low, medium, high dosage group of Yiliuning preparation are 34.09%,35.27%,38.85% respectively while the tumor inhibition ratios of Xeloda group,Xeloda combining with low, medium, high dosage group of Yiliuning preparation are 42.93%,44.95%,48.99%, 50.00%. the tumor inhibition ratios of low, medium, high dosage group of Yiliuning preparation and PYM group are 32.19% Yiliuning 35.31%,38.64%,44.95%, respectively while the tumor inhibition ratios of PYM combining with low, medium, high dosage group of Yiliuning preparation are 37.88%,47.98%,51.01%. The tumor inhibition ratios of low,medium,high dosage group of Yiliuning preparation and vp-16 group are 32.09%,35.74%,38.71%,48.57% respectively while the tumor inhibition ratios of vp-16 combining with low,medium,high dosage group of Yiliuning preparation are 47.14%,50.00%,51.43%.(The specific datas and statistical analysis of each stages can be seen in the results of the article)Conclusion:Positive control group and different dosages of Yiliuning preparation of each stage can inhibit s 180 tumor growth in mice. the size and the weight of transplanted tumor in other 7 groups are lower than the control group. Among these experiments,the tumor inhibition ratio of this group is outstanding in which chemotherapy drugs are combined with different dosages of Yiliuning preparation. The efficacy of tumor inhibition ratio of CTXcombining with Yiliuning preparation is the highest,followed by DDP combining with Yiliuning preparation,then VP-16 combining with Yiliuning preparation. PYM group with Xeloda group is not conspicuous in synergism. About the perspective of Pharmacy,we can reach conclusions as follows:the mechanism of CTX combining with high dosage of Yiliuning preparation can be called synergy; the theory of DDP combining with high dose of Yiliuning preparation can be called additive effect; sensitizing effect can be explained the principle of VP-16,PYM and Xeloda combining with Yiliuning preparation respectively.Pharmacy evaluation,from low to high, are as follows:cyclophosphamide, Cisplatin, Etoposide, Pingyangmycin and Capecitabine...
