Purpose:1. To investigate the difference between XWLC-05cells and A549cells.2. To investigate the effect of LY294002on the radiosensitivity between XWLC-05cells and A549cells, and to detecte cell cycle distribution and apoptosis.Materials and methods:1. The clonogenic assay was performed to determine the survival fraction. The parameters D0,Dq,SF2and N for the single-hit multitarget model and the parameters α,β,SF2for the linear-quadratic model were calculated.2. While the clonogenic assay, in all the doses of radiation A549cells and take some XWLC-05cells, LY294002processing A549cells and XWLC-05respectively cultivate the cells for0,12,24,48, and72hours, making single-celled levitation liquid, and analyze cell cycle distribution and apoptosis index by Flow Cytometric Analyses and TUNEL.Results:1. The clonogenic assay showed that:With the increase of the radiation dose, the survival number of A549cells and XWLC-05cells decreased, and, after adding LY294002inhibitors, the survival number of both A549cells and XWLC-05cells is lower than before. Under the same radiation doses, the survival number of XWLC-05cells is lower than that of A549cells.2. Flow Cytometric analyses showed that irradiation induced G2/M phase arrest in A549cells and XWLC-05cells in a dose-dependent manner. In A549cells, LY294002co-treatment with irradiation slightly enhanced G2/M arrest compared to control. In XWLC-05cells, LY294002evidently enhanced IR-induced accumulation of cells in G2/M phase.3. TUNEL assay demonstrates that the amount of apoptotic cells of both A549cells and XWLC-05cells increased when radiation dose increases. However, when irradiation and LY294002were combined, much more apoptotic cells were observed compared to IR or LY294002alone. But XWLC-05cell apoptosis rate is higher.Conclusions:1. LY294002could enhance radiosensitivity in A549cells and XWLC-05cells.2.There were differences in cellular intrinsic radiosensitivity between A549cells and XWLC-05cells.3. Simple drugs that inhibit the PI3K/Akt pathway on the G2/M phase arrest was not obvious, the only joint irradiation can cause cell cycle arrest, so an increase in apoptosis rate.
