Flickingeria fimbriata (B1.) Hawkes is distributed in southern China,including Hainan, Guangxi, and Yunnan Provinces. Its whole herb, which isused as the tonic and antipyretic agent in China, has been recorded in theGuangdong Chinese Materia Medica Standards. Few chemical research aboutthis plant was reported, and thus no quality standard has been recorded. In thisthesis, we investigated its chemical constituents systematically, and alsoestablished its quality standard, which might provide scientific basis for itsclinical application and further research. The results are as follows:1. From the EtOAc fraction of F. fimbriata,19chemical constituentswere isolated by various chromatographic methods, and the structures of14compounds were determined by analysis of spectroscopic data andphysico-chemical properties as followed: nine diterpenes:(9S,16R)-9,16,17,19-tetrol-ent-kaur-3-one(FJD-1),(6R,16R)-6,16,17,19-tetrol-ent-kaur-3-one(FJD-3),(11R,16R)-11,16,17,19-tetrol-ent-kaur-3-one(FJD-4),(1S,6R,16R)-1,6,16,17–tetrol-ent-kaur-3-one(FJD-9),(16R)-17-[(β-D-glucopyranosyl)oxy]-16-hydroxy-ent-kaur-3-one-19-yl veratrate(FJD-14),(16R)-17-[(β-D-glucopyranosyl)oxy]-16-hydroxy-ent-kaur-3-one-19-yl3,4,5-trimethoxybenzoate(FJD-15),(16R)-17-[(β-D-glucopyranosyl)oxy]-16-hydroxy-ent-kaur-1-en-3-one-19-yl cinnamate(FJD-16),(3R,16R)-17-[(β-D-glucopyranosyl)oxy]-3,16-dihydroxy-ent-kaure-19-yl cinnamate(FJD-17),(1S,16R)-17-[(β- D-glucopyranosyl)oxy]-1,16-dihydroxy-ent-kaur-3-one-19-yl cinnamate(FJD-21); one monoterpene:(3S,5R,6R,7E,9R)-3,5,6,9-tetrahydroxy-7-megastigmane(FJD-11); one flavonoid: vitexin2″-O-β-D-glucopyranoside(F1); one sterol: β-daucosterol(F2); two phenolic acid:3,4,5-trimethoxybenzoic acid(FJD-5), methyl3,4-dihydroxybenzoate(FJD-7). FJD-1,3,4,9,14,15,16,17and21are new compounds, whereas FJD-7, FJD-11, F1andF2were isolated from the genus Flickingeria for the first time.2. The contents of water, ash, and the amounts of extraction weredetermined according to the China Pharmacopoeia2010Edition.Reversed-Phase High Performance Liquid Chromatography (RP-HPLC) wasapplied for quantitative determination of vitexin2″-O-β-D-glucopyranoside inF. fimbriata. The methodology was examined for precision, stability andreproducibility, and the results showed that the method established in our tiralwas in accordance with the technical requirements. Using this method, thevitexin2″-O-β-D-glucopyranoside's content of3batches of F. fimbriata wasdetermined. Finally, we draw up draft standard of F. fimbriata.
