Objective: In order to observe the effect of yiqiyangyinhuoxue therapyon large vascular endothelail tissue,the expression of in vascular cell adhesionmolecule (VCAM-1) in diabetic rats,at the same time, to discuss theprecaution and treatment of the therapy and probalble mechanism on largevascular endothelail tissue, we established this diabetic rats model andexerted medicine intervention.Methods:46clean grade male of SD rats were selected with the weightfrom160g to180g.After one week, select randomly10rats as the normalcontrol(N) group, According to reference documents, apart from the normalgroup, the other rats were injected streptozotocin (STZ) in abdominalcavity, which was dissolved into1%buffer solution of citric acid of STZaccording to60mg/kg,and the normal group were injected with thecorresponding1%citric acid-sodium citrate.72hours later, detected ratsblood glucose. We regarded the rats whose numerical value of the bloodglucose more than16.7mmol/L.Then they were randomly divided into modelcontrol group(M), yiqiyangyinhuoxue (Z)group(called traditional Chinesemedicine group for short), simvastatin(X) group (called Western medicinegroup for short),12rats in each group. Then, the drugs were given to the rats,according to adult dosage10times,The rats of Z group were given theyiqiyangyinhuayutongmaifang, according to31g/kg·d, the X group were giventhe simvastatin, according to1.7mg/kg·d.Both Z group and X group weregiven drugs once aday, The N group and the M group were given thecorresponding quantity's physiological saline by gavage. After given themedicine for12weeks., All of the rats were forbidden to eat and drink for12h.Moreover they were anesthetized by injecting2%napental into abdomen,at the same time, all rat were executed femoral artery blood,separation of serum,detectd rats blood glucose, total cholesterol(TC), triglyeride(TG),lowdensity lipoprotein(LDL),and high density lipoprotein(HDL). get the specimen,and observe the change of pathomorphism of large vascular endothelail underlight microscope. The immunohistochemical method was used to detect theexpression of vascular cell adhesion molecule (VCAM-1)in large vascularendothelail.Results:1The general state in each of the groupsThe normal rats had not abnormal appearance in mentalstates,diet,drinking water,daily activities.But the modle group showedIndifference, polydipsia, polyphagia polyuria, emaciation, color dim,lassitude and retardation. Some rats appeared hemiparalysis, limbs edema.yiqiyangyinhuoxue group alleviated than of the model group.2The change of blood glucose and body weight in of the groupsBlood glucose levels of diabetic rats were significant higher than thenormal group after modeling after injected streptozotocin (p <0.01), but theWestern medicine group was not of significant differences compared with themodel group (P>0.05), In treatment groups, The blood glucose levels ofChinese medicine treatment group was significantly lower than that of modelgroup(P<0.01).Compared with the normal group, the body weight of the model groupand two treatment groups decreased significantly (P<0.01).The traditionalChinese medicine group in the body weight significantly increased (P<0.01)than the model group,but the Western medicine groupwas not of significantdifference compared with the model group (P>0.05)3The cholesterol (TC) and low density lipoprotein(LDL)of rats in eachgroup comparedDuring the experiment, TC and LDL in Model group was significantlyhigher than the normal group (P <0.01), Compared with the model group, Thetwo treatment groups were lower significantly (P<0.01), Western medicinetreatment group significantly decreaced than Chinese medicine treatment group(P<0.01).4The pathomorphological observation of large vascular endothelialmorphology in each groupWe can observed by light microscope after HE that normal cell textureclear and take the normal shape, color uniform cytoplasm, flat arteryendothelial cells, endometrial flat, smooth muscle cells(SMC) and elasticparallel plate order. Model rats' artery wall thickening of the endometriumappeared limitations, the smooth muscle cells in the film shallow proliferationmild, arranged in disorder, with lesions department interwoven arrangement,separation fracture. Rats' artery vascular endothelial morphology of theChinese medicine group changed more lightly than the model group.5The expression of vascular cell adhesion molecule (VCAM-1) in diabeticrats' large vascularThe expression of the vascular cell adhesion molecule (VCAM-1) indiabetic rats' thoracic aortic endothelial cells was significantly higher than thenormal group (P <0.01). compared with the model group, the expression of theChinese medicine group, VCAM-1was reduced (P<0.05), the difference wasstatistically significant. Chinese medicine treatment group's VCAM-1expression was lower than Western medicine treatment group (P<0.05)Conclusions:1YiQiyangyinhuoxuefang can improve general status of diabetic rats,reducethe blood glucose and blood fat, regulate glycometabolism and LipidMetabolism in diabetic rats.2YiQiyangyinhuoxuefang can improve pathologic morphous of diabetic rats'large vascular endothelial, protect the large vascular endothelial meanwhile,and further provid a feasible treatment Protocols in clinical application3The effect of Prevention and cure on diabetic macroangiopathymaily beowing to its improvement of the general status,the blood glucose and theexpression levels of VCAM-1.
