Effect Of "YiQiYangYinHuoXue" Therapy On ET-1in The Diabetic Rats’ Large Vascular

Objective: YiQiYangYinHuoXue recipe is the Chinese medicine formulathat my teacher built according to clinical practice in the treatment of diabeticmacroangiopathy. This experiment established diabetic rats model bystreptozotocin, then apply the YiQiYangYinHuoXue recipe to the diabetic rats,and observed the diabetic rats’ changes on general behavior, weight, glucose,blood lipid, blood vessel endothelium form and the endothelin1(ET-1), thendiscuss the possible mechanism on prevention and cure diabeticmacroangiopathy by using this prescription.Methods: We choose sixty male clean level Wistar rats, at the weight of180g～220g, and let them adapt the environment for one week, during thattime, we provide enough food and water. After that, randomly select15rats asnormal group (N), other rats were injected streptozotocin(STZ) in abdominalaccording to55mg/kg, which was dissolved into1%buffer solution ofcitricacid of STZ. The normal group were given corresponding1%citric acid,sodium citrate solution injection as the same volume as the model bulit group.After72hours, test the glucose from the tail blood, if the glucose more than16.7mmol/L, and the rats drink water quantity, eat more food and urine outputincreased significantly, the diabetic rats were successfully made. Thenrandomly divided the diabetic rats into three groups, model group (M),Chinese traditional medicine group (Z) and simvastatin group (westernmedicine group, X), each group has15rats, then we use the Chinese medicineand simvastatin to treat on Z group and X group once per day, Z groupgavaged with YiQiYangYinHuoXue recipe by20g/kg per day. X group wasgiven simvastatin tablet2mg/kg per day. The N group and M group dailygiven the same volume of physiological saline, the gavage lasted12weeks.After final gavage, forbidden the rats’ eat food for12h, then injection2% sodium pentobarbital intraperitoneal, anesthesia, femoral artery take blood todetection of plasma glucose, cholesterol, triglyceride, low density lipoproteincholesterol and high-density lipoprotein cholesterol. Aseptic operation takethoracic aorta and put it in4%of paraformaldehyde solution, make specimens,use HE staining, and observe the change of vascular endothelial tissue,immunohistochemical to measuring thoracic aorta endothelium vascularendothelin1(ET-1), and observed the thoracic aorta endothelium pathologicalmorphology change.Results:1. General observationNormal group’s rats, act freely, eat food and drinking water as usual,normal urination. But in model group, the rats appeared listlessness, obviouslypolydipsia, polyphagia, polyuria, angular. Few withered and yellow color,burnout’s lie, slow response, etc. Some rats appeared mild hemiplegia, limbedema wait for a symptom. Chinese traditional medicine group’s rats theabnormal performance were less than the model group, western medicinegroup of rats in general are inferior to the traditional Chinese medicine groupof rats.2The changes of glucose and body weightCompared to the normal group, the glucose in model group and twotreatment groups were higher, the difference was significant (P<0.01);Western medicine group compared to the model group, the glucose didn’tchange a lot, there was no statistical significance (P>0.05); Chinese traditionalmedicine group compared with model group the glucose declined obviously,and the difference was statistically significant (P<0.01).Compared with the normal group, model group and the two treatmentgroups’ body weight dropped obviously, there was a statistical significance(P<0.01); Compared to model group, Chinese traditional medicine group’sbody weight increased significantly (P<0.01); but the western medicine groupcompared to the model group, the body weight increased no significantly(P>0.05). 3Comparison of blood lipid in each groupThe cholesterol (TC) and low density lipoprotein (LDL) in model groupand Chinese medicine group and western medicine group’s cholesterol (TC)were higher than normal group (P<0.01); The treatment group compared withmodel group, the total cholesterol (TC) and low density lipoprotein (LDL)were significantly lower, the difference was significant (P<0.01); comparedwith Chinese medicine group, the TC and LDL in Western medicine groupwas lower, the difference was significant (P<0.01).4Observation of pathological morphology on vascular endothelial tissue ineach groupAfter HE staining, normal group cells display texture clear, shape wasnormal, cell cytoplasm coloring uniformity, aortic endothelial cells lining flat,smooth, in the film smooth muscle cell (smooth muscle cell, SMC) and elasticplate and present a parallel arrangement. Model group of rat aortic intimathickening of the limitations, endometrial local protuberance, membraneshallow of smooth muscle cells visible hyperplasia, arrangement disorder,elastic plate arrangement irregular wavy, rarely appear rupture, separation.Chinese traditional medicine group’s form of organization was better than themodel group. But the western medicine group’s form was not good enough.5Expression of endothelin1(ET-1) in rats’ vascular among each groupCompared with the normal group, model group and the treatment grouprats thoracic aorta vascular endothelin1(ET-1) is obviously higher (P<0.01).Traditional Chinese medicine group compare with model group comparison,the ET-1expression decreased (P<0.05), the difference was significant.Comparison between two treatment groups, Chinese traditional medicinegroup the ET-1reduce significantly, the difference was statistically significant(P<0.05).Conclusions:1The YiQiYangYinHuoXue recipe can improve the general condition ofthe diabetic rats, reduce glucose and blood lipid. It also had the protectionmacroangiopathy positive effects, and inhibits endothelial element1secretion. 2The YiQiYangYinHuoXue recipe can improve diabetes rats’ vascularendothelial tissue pathological form, and protect large vascular endothelialfunction, thus provide a possible treatment to clinical application.
