Screening Of Anti-oxidative Strains And Its Anti-aging Study On D-galactose Mice Model

The objective of the paper was to study a series of antioxidative intact cells and their products.First of all,4lactic acid bacteria and6yeasts were screened by scavenging1,1-diphenyl-2-picrydrazl (DPPH) free radical and hydroxyl radical from isolated26strains in different food,and superocide radical scavenging ability, reducing power and inhabiting lipid peroxidation were further exprimentized. Second, Second,The main ingredients in these strains and their products were analyzed on the antioxidant and used to Follow-up experiments.At last,aged model mice which were given subcutaneous injection of D-gal continuously, were fed with intact cells and their products,and some anti-aging index were texted. The results are as followed.The results showed the DPPH free radical,hydroxyl radical,superocide radical scavenging ability, reducing power and inhabiting lipid peroxidation by viable cells(L1,L2,Y3,Y5) and their products were hign obviously.In the polysaccharide content,the highest content of fermentation broth of L1, L2,Y3,Y5were124.0ug/mL(10h),112.4ug/mL (6h),188.5ug/mL (27h),345.1ug/mL of(21h) at different times,and Y3was the top.In Vc content,four strains was very low and negligible.In the flavonoid content,of which L1,L2,Y3,Y5in fermentation brothes were68.86ug/mL,45.06ug/mL,10.24ug/mL,5.12ug/mL,higner than their broken liquid content.In the Polyphenol content,of which L1,L2in broken liquids content was further higner than fermentation broth content,Y3,Y5were on the contrary.In SOD activity,of which L1,L2,Y3,Y5in fermentation brothes were lower than broken liquids,of which Y3was the hignest.Therefore,the subjects from Y3,Y5broken liquids and L1,Y3fermented brothes were used to feed mouse.To explore the optimal dosage of D-galactose for inducing aging mice model,and two hign-dose group mouse have died during the experiment,so the middle dose was chose as the optimal dose(650mg/kg)and injection time was6weeks.In the mouse weight gain and organ indices,L1fermentation broth group, the Y5bacterial group,Y3fermentation broth group,Y3bacterial group growth and weight gain in mice were7.7g,8.1g,9.9g,8.7g,10.0g,the other organs of moder group is lower than average group except heart and kidney.Antioxidant enzymes(SOD, POD, CAT,GSH-PX),T-AOC,MAO and MDA content of the mice induced with D-galactose were changed in some extent.The different subjects for6weeks can change the indicators of indexes but there are different effects on indexes.
