Objective:This paper aims to improve the method of constructing Qi deficiency animal model using Qi deficient mice, and to explore the effects of Shen Ling E Jiao Compound Ointment (SLEJCO) on three physiological functions, namely anti-fatigue, anti-oxidation and immunomodulation, in Qi deficient mice, and their relevant mechanisms.Methods:Qi deficient mouse model which meets the experimental standards is selected by comparing four modeling approaches of exhaustive swimming, water standing, composite model group â…  (free swimming combined with water standing) and composite model group â…¡ (diet control and free swimming combined with water standing) with TCM manifestations of Qi deficiency constitution as the theoretical basis by observing the behavior of mice, calculating their body weight and ratio of exhaustive swimming time, and determining the physiological indices such as GLU, TC, TG, HDL-C, LDL-C, hepatic glycogen content, BUN level and LDH activity. SLEJCO, as the intervening agent, is administrated intragastrically at high-medium-and low-doses of 10.5 g/kg,21.0 g/kg and 42.0 g/kg (crude drug/mouse body weight); and an equivalent volume of normal saline is intragastrically administered to mice in the normal control and Qi deficiency model groups.19 days after continuous administration, behavioral differences among mice in various groups are observed, and body weight, ratio of exhaustive swimming time, GLU, TC, TG, HDL-C, LDL-C, hepatic glycogen, SUN level, LDH, SOD, MDA, GSH-Px activity, spleen index and thymus index of mice in each group are determined. Experimental data are processed using SPSS 19.0 statistical software.Results:Compared with the normal control group, mice in the Qi deficiency model group had reduced activity, and showed fatigue behaviors. Their body weight, exhaustive swimming time ratio, hepatic glycogen content, SUN level and LDH activity decreased significantly (P<0.05), while GLU was significantly elevated (P<0.05). After intervention with SLEJCO for 19 days,body weight, exhaustive swimming time ratio, hepatic glycogen content, LDH activity, TG, TC, HDL-C and LDL-C of mice in the SLEJCO low-dose group showed no significant differences from the normal control group (P>0.05), while GLU, BUN level, SOD activity, GSH-Px activity, thymus and spleen indices were significantly higher than the normal control group (P<0.05), and MDA activity was significantly lower than the normal control group (P<0.05). In the SLEJCO medium-and high-dose groups, body weight, GLU, TG, TC, HDL-C, LDL-C and BUN level had no significant differences compared with the normal control group (P>0.05), while exhaustive swimming time ratio, hepatic glycogen content, BUN level, LDH activity, SOD activity, GSH-Px activity, thymus and spleen indices were significantly higher than the normal control group (P<0.05), and MDA activity was significantly lower than the normal control group (P<0.05).Conculsions:1.By a combination of diet control, exhaustive swimming and water standing, Qi deficiency constitution resulted from weakened spleen and stomach functions can be well simulated, which is a quite ideal modeling approach.2. As specific physiological indices for evaluating Qi deficiency constitution, blood glucose, serum urea nitrogen (SUN), hepatic glycogen and lactate dehydrogenase (LDH) can well reflect the bodily fatigue and immunocompromise caused by glucose and protein metabolism disorders, which eventually lead to Qi deficiency constitution.3. SLEJCO made in accordance with TCM theory has a role in reducing the blood glucose, SUN and serum MDA levels of Qi deficient mice; while playing a role in elevating the exhaustive swimming time ratio, LDH activity, SOD activity, GSH-Px activity, spleen index and thymus index. Such elevations and reductions suggest that SLEJCO can enhance the anti-fatigue, anti-oxidation and immunomodulatory functions of Qi deficient mice. This lays the theoretical foundation for its market introduction.
