Objective:To explore the inhibition mechanism of moxibustion and moxa-smog on acute gastric mucosa damage through observing the influence of moxibustion and moxa-smog on acute gastric mucosa (AGML) rats with thier stomach tissues and morphological changes, content changes of antioxidant factors SOD,MDA in vivo, content of cell apoptosis factors BCL-2,BAX and AI; To compare the restoration functions of moxibustion with smog, moxibustion without smog and moxa-smog on acute gastric mucosal injury through analysing the protective effect of acute gastric mucosal injury in rats by the above three different methods.Methods:40SPF SD rats were randomly divided into5groups, that was A for normal control group, B for model group, C for smog moxibustion group, D for non-smog moxibustion group, E for moxa-smog therapy group, making acute gastric mucosa damage model in the way of filling anhydrous alcohol in the stomach. Observing gastric mucosa tissue morphological changes by HE staining in light-microscopy, pathological scoring on inflammatory reaction level, detecting SOD, MDA content by colorimetric method, detecting gastric mucosa BAX, BCL-2changes in rats by immunohistochemical method, detecting cell apoptosis index in rats of gastric mucosa by TUNEL method.Results:1,Stomach tissue morphological changes in rats:stomach tissue in normal control group had integrated structure without any damage. Compared with normal control group, gastric mucous membrane had sufferedc significant necrosis, inflammatory cells infiltration, and acute inflammatory response with remarkable hyperemia in model group; compared with model group,, the surface of gastric mucosal had partly necrosis, obviously improved intravascular congestion conditions, less inflammatory cells infiltration in smog moxibustion group and non-smog moxibustion group. Compared with smog moxibustion group and non-smog moxibustion group, the surface of gastric mucosal had serious damage, more apparent inflammatory cells infiltration, and intravascular congestion in blood vessels in moxa-smog group.2,Comparison of pathological scoring on inflammatory reaction in gastric mucosal tissue in all groups:compared with normal control group, pathological scoring in model group rised significantly (P<0.01); pathological scoring in smog moxibustion group and non-smog moxibustion group were significant lower than in model group (P<0.01); pathological scoring on inflammatory reaction in gastric mucosal tissue in smog moxibustion group was significant lower than in moxa-smog group and there was significant difference (P<0.01); pathological scoring in Smog moxibustion group and non-smog moxibustion group were no significant difference; there were no significant difference between pathological scoring in moxa-smog group and model group.3,Comparison of SOD of blood serum in all goups:compared with normal control group, the value of SOD in model group reduced significantly (P<0.01); compared with model group, the value of SOD in smog moxibustion group and non-smog moxibustion increased significantly, and showed significant difference (P<0.05); the value of SOD in smog moxibustion group was significantly higher than in moxa-smog group and there was significant difference (P<0.05); the value of SOD in Smog moxibustion was a little bit lower than in non-smog moxibustion group, but there were no significant differences between the two groups; the value of SOD in moxa-smog group was a little bit higher than in model group, but there were no significant differences between the two groups.4,Comparison of MDA of blood serum in all goups:compared with normal control group, the value of MDA in model group rised significantly (P<0.01); compared with model group, the value of MDA in smog moxibustion group and non-smog moxibustion reduced significantly, and showed significant difference (P<0.01); the value of MDA in smog moxibustion group was significantly lower than in moxa-smog group and there was significant difference between them (P <0.01); the value of MDA in Smog moxibustion was a little bit lower than in non-smog moxibustion group, but there were no significant differences between the two groups; the value of MDA in moxa-smog group was a little bit lower than in model group, but there were no significant differences between the two groups.5,Comparison of the average grey values of BCL-2in gastric mucosal tissue in all groups:compared with normal control group, average grey value of BCL-2in model group rised significantly (P<0.01); compared with model group, average grey value of BCL-2in smog moxibustion group and non-smog moxibustion group reduced significantly, and showed significant difference (P<0.01); the average grey value of BCL-2in smog moxibustion group was significantly lower than in moxa-smog group and there was significant difference between them (P <0.01); expression of BCL-2in Smog moxibustion group and non-smog moxibustion group were no significant difference; there were no significant difference between average gray values of BCL-2in moxa-smog group and model group.6,Comparison of the average grey values of BAX in gastric mucosal tissue in all groups:compared with normal control group, average grey value of BAX in model group reduced significantly (P<0.01); compared with model group, average grey value of BAX in smog moxibustion group and non-smog moxibustion group rised significantly, and showed significant difference (P <0.01); the average grey value of BAX in smog moxibustion group was significantly higher than in moxa-smog group and there was significant difference (P<0.01); expression of BAX in Smog moxibustion group and non-smog moxibustion group were no significant difference; there were nosignificant difference between average gray values of BAX in moxa-smog group and model group.7,Comparison of cell apoptotic index in gastric mucosal tissue in all groups:compared with normal control group, apoptotic index in model group rised significantly (P<0.01); apoptotic index in smog moxibustion group and non-smog moxibustion group were significant lower than in model group (P <0.01); the cell apoptotic index in gastric mucosal tissue in smog moxibustion group was significantly lower than in moxa-smog group and there was significant difference(P<0.01); apoptotic index in Smog moxibustion group and non-smog moxibustion group were no significant difference; there were no significant difference between apoptotic index in moxa-smog group and model group. Conclusion:1. Smog moxibustion and non-smog moxibustion had protection forgastric mucous membrane from gastric mucosal injury in rats,and there were no obvious differences between the two groups.2. Smog moxibustion therapy compared with non-smog the antioxidant and cell apoptosis effect has apparent difference, Smog moxibustion effect better.3. Moxa-smog on the acute injury of gastric mucosa protective effect is not obvious, and compared with acute gastric mucosa injury self protection restoration effect has no significant difference.4. Smog moxibustion, non-smog on acute gastric mucosal injury has a protective effect.the protection mechanism might be that promoting gastric mucosal tissue to repair by raising expression of BCL-2and reducing expression of BAX in order to regulate dynamic homeostasis in the content of BCL-2and BAX; by raising the content of SOD and reducing the content of MDA, in order to enhance antioxidant effect and reduce inflammation response.
