Objective:This project investigated the effects of moxibustion pretreatment on the expression of TFF and PAR in gastric mucosa of stress gastric ulcer rats.To explore the mechanism of moxibustion promoting gastric mucosal lesion repair,so as to provide experimental basis and theoretical support for moxibustion treatment of gastric ulcer diseases.Materials and Methods: 60 healthy male SPF SD rats were randomly divided into blank group,model group,moxibustion Group,moxibustion control group and Omeprazole Group,and 12 rats per group.The experiment was designed to keep the rats in a blank group,model group and omeprazole group from the first day to the eighth day,and to carry out the same binding and fixation for 30 minutes a day for these rats.At the same time,the moxibustion group and the moxibustion control group were not only required to bind and fix the rats,but also to the corresponding acupoints of these rats moxibustion for 30 minutes.In addition,on the eighth day of the experiment,after the rats in each experimental group were treated with experimental factors,rats in each group were fasting for 24 hours.In the ninth day of the experiment,the model was made with ahydrous ethanol irrigation stomach's method.In the 30 minutes before the model was made,2ml/100 g omeprazole was injected intraperitoneally in the rats of omeprazole group,and 2ml/100 g 0.9% sodium chloride solution was injected into the abdominal cavity of the other groups' s rats.Then,except for the blank group's rats,the rest were infused with 0.5 ml/100 g doses of anhydrous ethanol.The rats in the blank group were subjected to 0.9% sodium chloride solution with 0.5ml /100 g standard.After the model was finished,the rats' s gastric mucosal blood flow was monitored by laser speckle blood flow imaging technique after anesthesia.The damage index of gastric mucosa in rats was calculated by GUTH method.The morphological changes of gastric mucosa tissue in rats were observed under light microscope.The content of TFF in gastric mucosa tissue of rats was detected by enzyme-linked immunosorbent assay(ELISA).The expression of PAR in gastric mucosa tissue of rats was detected by immunohistochemical method(IHC).Results: 1?Compared with the blank group,the gastric mucosal blood flow of rats in model group was much lower,and the change was statistically significant(p<0.01).Compared with the model group,the gastric mucosal blood flow of the rats in moxibustion group,moxibustion control group and omeprazole group had a significant increase,which was statistically significant(p<0.01).Moreover,the gastric mucosal blood flow of the rats in moxibustion group was significantly higher than that of moxibustion control group,and the change was statistically significant(p<0.01).2?Compared with the blank group,the damage index of gastric mucosa of rats in the model group was significantly increased,and the difference was statistically significant(P<0.01).Compared with the model group,the injury index of gastric mucosa of the rats in moxibustion group,moxibustion control group and omeprazole group had a significant increase,which was statistically significant(p<0.01).Compared with moxibustion control group,the gastric mucosal injury index of the rats in moxibustion group decreased significantly,Compared with the moxibustion control group,the decrease of gastric mucosal injury index in the moxibustion group was more significant,and the difference was statistically significant.(p<0.01).3?Compared with the blank group,the gastric mucosal cells of rats in the model group were vague,disordered,and had significant cell infiltration,as well as a large number of necrotic cells were observed.Compared with the model group,The mucosal injury of the rats in the moxibustion group was relatively light.In terms of cell alignment,the cells are ordered and most cells are intact.and moxibustion control group of rats damage degree than moxibustion group serious,gastric mucosal cell arrangement is also irregular,distribution is not regular,showing complex phenomenon.Occasionally,there are cell infiltration and cell edema.4?Compared with the blank group,the contents of TFF in the gastric mucosa of rats in the remaining groups increased markedly,and the changes were statistically significant(p<0.01).Compared with the model group,the content of TFF in gastric mucosa of rats in moxibustion group,moxibustion control group and omeprazole group increased significantly,and their differences were statistically significant(p<0.01).Compared with moxibustion control group,the content of TFF in the gastric mucosa of the rats in the moxibustion group increased more significantly,and the difference was statistically significant(p<0.01).5?Compared with the blank group,The expression of PAR protein in the gastric mucosa of rats in the remaining group was elevated,and the difference was statistically significant(p<0.01).Compared with the model group,the expression of par protein in gastric mucosa of rats in moxibustion group,moxibustion control group and omeprazole group increased in different degrees,and this difference was statistically significant(p<0.01),Moreover,the moxibustion group was significantly higher than the moxibustion control group(P<0.01).Conclusion: 1.Moxibustion has a certain preventive and protective effect on gastric mucosal injury induced by anhydrous ethanol perfusion,and the effect of moxibustion group is better than that of moxibustion control group.2.Moxibustion can increase the blood flow of gastric mucosa and promote the repair process of gastric mucosal lesion.3.Moxibustion can increase the content of TFF and PAR in the gastric mucosa of rats,thus promoting the proliferation and repair of gastric mucosal lesions.
