The Cloning And Functional Analysis Of Interleukin 1 Receptor Antagonist, SIL1rn And Mus Musculus Glutathione Peroxidase 7, GPx7-Two Hematopoietic Associated Cytokines

The interleukin (IL) superfamily is a group of important cytokine. During our research we have found that sILlrn (secreted interleukin 1 receptor antagonist) have the effect of positive regulatory cytokine, for HSCs (Hematopoietic Stem Cells) expansion.First we establish the 5-FU mode of mouse by injection of 5-FU (5-Fluorouracil.), displaying the repression and regeneration process of mouse bone marrow. By cDNA Microarray and data analysis, we have studied in the whole process of this modle the different level of gene expression at day 0, 3, 7, 11, and 14 respectively, and have also got the gene exprssion spectrum of the modle. On the basis of this spectrum, we have found a group of cytokines that play important roles in association with positive hematopoietic regulation. Presented in this paper we cloned two of these genes, sILlm and Gpx7, and subcloned them into the the eukaryotic expression vector phCMV and pcDNA3.1, and got high expression of this gene in mouse TA (Tiabials Anterior) muscle via muscle injection, electroporation, and in vivo expression. The result turn to be, the in vivo expression of sILlrn could significantly increase white blood cell numbers of peripheral blood of the modle mouse treated with 5-FU, it could decrease BM repression time from 14 days to 11 days. And the expression of Gpx7 could significantly increase bone marrow cell numbers. At the same time, growth hormone (GH) was discovered have cooperative effect with sILlm, We primarily studied the function by injection of fusion GH protein via s.c.
