Candida albicans has been the most frequently isolated fungus in immunocompromised patients associated with mucosal and deep-tissue infections. We rapidly constructed a full-length cDNA library of C. albicans using switching mechanism at 5' end of RNA transcript (SMART) techniques, and prepared the Cadida albicans cDNA microarray consisted of 3096 unigenes we have got. In the present study, we examined the large-scale gene expression profiles with six sets of matched clinical and experimental fluconazole susceptible and resistant C. albicans strains by microarray analysis. We observed that azole resistances developed in multiple circumstances are all associated with increased expression of CDR mRNA. We also identified some common differential expressed genes and some interesting function-linked gene groups involved in drug-resistance. Comparing the differnences between the resistant strains and sensitive strains, we have screened some novel genes and got their complete coding sequences rapidly. Based on the analysis of bioinformatics, two novel genes CaFDH12 and CaHXTlO were chosen to be made a systemic evaluation of their roles on drug sensitivity, transport, cell cycles, morphology and virulence in C. albicans.
