Water environment pollution mainly involves point source pollution and non-pointsource pollution, Governments around the world pay attention on the non-point sourcepollution, Western Europe and the United States of some developed countries carry outlarge number of experiments to achieve effective control and management on non-pointsource pollution. And for the non-point source pollution rarely research was reported,non-point source pollution in the United States in the legislation is defined as with thespread of pollutants wap trace form into the waters of surface and underground.Compared with the point source pollution non-point pollution has the characteristics ofConcerned range, Hidden Strong, randomness, long Latent cycle, Cumulativehigh.Making it difficult to monitor, difficult to quantify. presently it has become themain affect water environment quality pollution sources, Therefore seeking an effectivemonitoring, non-point source pollution technology has become the social pollutionsources research hot spotMST (Microbial Source Tracking) technology was a new technology developed inthe mid1990s, which was called a technology which can change non-point Sourcepollution monitoring into point Source pollution monitoring. This paper was fornon-point pollution on the feces pollution. After consulting a large number of materialand the many experiments, the HF183F-Bac708R and Biado1-Biado2was selected ashuman excrement specific gene markers primers, using DF475F-Bac708R as the dogfeces specific genetic markers primers, respectively established methods fromextracting DNA to the PCR detection methods of qualitative and quantitative. Thebacterial genomic DNA extracted kit was choosed to extract the total bacterial DNA ofenvironment water, through the temperature gradient function of PCR detection system and the annealing temperature optimization to improve PCR detection effect and themethod of gene markers was confirmed.Specific gene markers quantitative PCR detection methods were established basedon the qualitative detection method.This study also used recombinant plasmid and thegenomic DNA of standard strain as templates to create standard curves, respectively,and then the two kinds of standard curve were compared. Furthermore, this researchused the polymerase chain reaction (PCR) established simulating natural conditions, theinfluence of different temperature and without illumination of genetic markers livecharacteristics was researched, In addition a simple analysis of genetic markers forremoving was made on different processing unit of the first and the third sewagetreatment plant of xian city sewage.
