Probiotic dietary supplements have the efficacy of maintain bowelfunction; preventing constipation and strengthening the immune system.It’s predicted that the global value of probiotic dietary supplements canreach to$1.7billion. This research aimed to successfully prepareprobiotic dietary supplements which rich in highly active probiotic andcontained apple pomace nutrients, with the use of apple pomace,Lactobacillus acidophilus, Lactobacillus bulgaricus and Streptococcusthermophilus as fermentation bacteria. The results are as follows:Single strain fermented apple pomace extract experiment results:First by single-factor test screened three kinds of probiotics LA, LB,ST fermented apple pomace extract want to add nutrients added and theaddition amount. Then, the ratio of apple pomace and water, extractiontemperature and extraction time was the single factor screening the bestextraction conditions. Last, the optimal fermentation conditions weredetermined by response surface methodology(RSM) analysis as follows:the ratio of apple pomace and water was1:8~1:10,90~100℃extracted50~60min, filtered and had the extraction solution,Lactobacillus acidophilus LA added1.06%edible yeast extract,1.54%Whey Powder,37℃fermented28.14h, the number of viable cellsreached5.24×109cfu/ml; Lactobacillus bulgaricus LB added1.55%edible yeast extract,2.2%Whey Powder,37℃fermented49.24h, thenumber of viable cells reached2.51×109cfu/ml; Streptococcusthermophilus ST added2.12%edible yeast extract,2.65%Whey Powder,41℃fermented44.86h, the number of viable cells reached4.93×109cfu/ml.LA, LB, ST mixed strain fermented apple pomace extractexperiment results: On the basis of the single strain fermentation test results, the optimal conditions of mixed strain fermented apple pomaceextract were determined by mono-factor experiment and responsesurface methodology (RSM) analysis as follows: the ratio of apple freshslag and water was1:8~1:10,90~100℃extracted50~60min, filteredand added1.97%Whey Powder,1.43%edible yeast extract,115℃sterilized20min, then, the inoculation ratio of LA, LB, ST was1:2:1,total inoculation volume was4%,39℃fermented30.67h, the number ofviable cells reached4.78×109cfu/ml.Results of semi-solid state fermentation of apple pomace: First,determined the best material to water ratio and the best beating time bybeating experiment. Then, by the test of adding whey powder andcontinuing to add the edible yeast extract, determined the best formula.Last, the best fermentation process was: fresh apple residue with waterby a ratio of1:5beating6min, added1.5%whey powder,2%edibleyeast extract,115℃sterilized20min, then, the inoculation ratio of LA,LB, ST was1:2:1and total inoculation volume was4%,39℃fermented48h, the number of viable cells reached8.87×108cfu/ml.Preparation of liquid probiotic dietary supplement experimentresults: By the addition of dietary fiber powder, sucrose, citric acid andstability test, determined the best formula: Highly active probioticbacteria composite dietary fiber powder1.25%, added6%sucrose,0.1%citric acid and0.2%sodium carboxymethyl cellulose(CMC-Na), thengot the dietary supplements rich in dietary fiber and highly activeprobiotic. Its color was yellow, and no stratification, no precipitation,uniform texture, good fluidity, suitable for sweet and sour, soft anddelicate taste. The number of probiotic viable cells was4.05×109cfu/ml.Every100ml of finished products contains crude protein15.16g anddietary fiber1.23g.Preparation of Semi-solid probiotic dietary supplements results:By the test of add sucrose, citric acid and sodium carboxymethylcellulose(CMC-Na), determined the best formula: After fermentation,highly active probiotic jam added8%sucrose,0.15%citric acid and0.5%CMC-Na. The color of finished product was pale yellow, shiny, glue–like with a certain mobility, but not dispersed and not the flow ofjuice, not sugar-free crystalline, sweet and sour suitable for delicate andrefreshing. The number of probiotic viable cells was8.65×108cfu/ml.Every100g of crude protein content in the finished product was21.15g,and dietary fiber was1.04g.
