Paederia scandens which is rubiaceae paederia scandens belongs to a kind of herbs. Its characteristics are used in food and medicine, widespread and cultivate a method simple. According to the concepts of traditional Chinese medicine, paederia scandens with root and whole herb medicine has the effects of relieving swelling and pain, relieving toxin and activating blood, promote digestion of the plot and dispel wind and eliminate dampness. It has been widely used in modern clinic medicine. Researches show that paederia scandens is also rich in ascorbic acid, flavonoids and a variety of biologically active materials,so it has some effects that are more adapted to Modern medical applications such as preventing cardiovascular disease, regulating Immune System, anti-aging and so on, its unique health effect is also recognized gradually by people. Therefore, the chemical composition, pharmacologi-cal effects and extracted method about active ingredient for paederia scandens have been paid more and more attention.This paper took an exploratory research to the technology of supercritical CO: extraction active ingredient from paederia scandens. By studying the extracting factors, the absolute ethylalcohol was determined as the entrainer. in the condition of1L extraction kettle and100g raw material, the amount of entrainer is500ml.Batch adding was qualified as the optimal injection modes, the influences of CO2flow rate and raw material particle size for extracting effects were overcome, the optimum technological parameters were obtained by changing the pressure, temperature and time.Using the extraction rate as an assessing index according to the uniform experiment design to examine the effect of factors on supercritical CO? extraction active ingredient from paederia scandens experiment respectively. In the selected level range. the affecting degree of factors sepence:extract pressure>extract time>extract temperature. Finally, the optimum process conditions were obtained:extract pressure32Mpa. extract time62℃.extract temperature1.75h.Based on this optimum process conditions, the actual maximum yield was8.84%. Through analyzing of the chemical constituents of supercritical carbon dioxide extraction products of paederia scandens by GC-MS technology,19compounds were identified, where Seven kinds of components such as N,N-Dimethylpropionamide,1-Cyclohexyldecane, L-ascorbyl-2,6-dipalmitate,1,2,3-propenetricarboxylic acid, tri(2-methylpropyl) ester, tributyl citrate, acetyl tributyl citrate, squalene, which haven’t been received via traditional extraction methods, were separated from paederia scandens. In these identified compounds, L-ascorbyl-2,6-dipalmitate, isoborneol, oleanolic acid, squalene have good therapeutic effects, and2-(Methylthio)benzaldehyde which contains sulfur is the possible component of the characteristic smell of paederia scandens.Comparing the reference standard of squalene, we carried on qualitative and quantitative analysis of the squalene contained in the chemical constituents of supercritical carbon dioxide extraction products of paederia scandens with gas chromatography. The analysis results show that the amount of squalene is proportional to the yield of extracts. On the optimum process conditions, the amount of squalene is maximum, the yield is about74mg/100g.
