With the development of modern sciences and technology and the improvement of social economy and human's living standards, however, "health" is still the most important topic in the world. Nature products are to be the hotspot in the world, more and more medical and food industries and more and more people pay more attention on which.Humulus Scandens (Lour.) Men. is Moraceae wildness liane, it can be found everywhere in China except Xinjiang and Qinghai. All parts of this plant can be used as medical materials. It is cool in nature, treating inflammation, tussis and so on, which has a long history of several hundred years in using as medical material in China. However, it is regarded as undesirable weed usually, because it's growth is very fast in a short time and may rob nutrition from crops. Few people have studied the plant. and what they studied were focused on medical functions of water extraction, not involving the systemic isolation and identification of components and purification related for main chemical components. Thus, in this paper, the selection of the factors for extraction, and detecting of the content of luteolin, especially isolation and purification luteolin were studied. In these experiments, several methods as Column chromatography, HPLC, HSCCC and UV-vis were used. All datas and results in this study are true, which not only provided a new method for developing other botanical resources, but also many theoretical and practical datas for developing.The results showed that:1. A RP-HPLC method for the detection and measurement of luteolin in Humulus Scandens (Lour.) Men. was developed firstly. Operating conditions were UV-350nm detection, allsphere OSD-25u column 250mm × 4.6mm i.d., and the flow phase of v (methanol) /v (water)=(50:50, water was adjusted to pH=3.0 with acetic acid). the flow speed of 1.0mL/min, 10μL injection volume, at room temperature for column. Luteolin contents were about 0.325mg/g in the young leaves and tender stem. All dates above identified that the plant is a very important botanical resource, which has wide developing future.2.The material powder were extracted by alcohol solution (80%) at 80℃ 60min, then adsorbed by polyamide chromatography column and eluted by 60% alcohol solution.3.The raw material of 19.1% Luteolin were purified into 98.5% effectively by High-Speed Countercurrent Chromatography (HSCCC). The separation conditions were determined as 800r/min rotated rate, 1.5ml/min flow rate, 50mg injection volume in the solvent system...
