This paper studied the extraction process conditions of lutein from corn protein powder by supercriticalCO2and Ultrasound method, the purity of lutein was determined by HPLC. The technology parameter of luteinextraction was studied by response surface analytic.The results showed that the optimum supercritical CO2extraction conditions were: extraction time is2.3h,temperature is42℃,pressure is30MPa,φ(ethanol)=90%asthe entrainer and the additive amount is5%.Under these optimum conditions, the purity of lutein is46.51%;and the ultrasonic-assisted extraction conditions were: φ (ethanol)=89%, extraction time is33min,temperature is39℃, solvent rate is15, ultrasonic power is strong,the purity of lutein is35.01%and issignificantly lower than that of supercritical CO2extraction,so it is choose the supercritical CO2extractionmethod as the optimum extraction method.This paper studied time,temperature and material-liquid ratio of the saponifition process conditions oflutein from corn protein powder by supercritical CO2,the purity of lutein was determined by HPLC.Thetechnology parameter of lutein saponifition was studied by response surface analytic. One-factor-at-a-time andresponse surface methods were used to optimize the conditions for the optimum saponifition processparameters using the purity of lutein as index and response value respectively.The results showed that theoptimum saponifition conditions were: saponifition temperature is52℃, time is4.7h, solid-liquid ratio is1:11,concentration of potassium hydroxide in ethanol is10%.Under these optimum conditions, the purity of luteinis67.38%and the verification is67.42%.The further isolation and purification of lutein by the method of C30column chromatography.The feedconcentration,elution agents proportion,elution agent,flow rate were investigated.The optimum processconditions was: the feed concentration was20mg mL-1,the sample solvent was hexane,the temperature was30℃, the eluent was V(ethanol):V(MTBE)=97:3,the eluate volume was4BV,the flow rate is2.0mL min-1, andthe purity of lutein can be increased from67.42%to95.37%.
