| Ibuprofen is the most effective Non-steroidal analgesic andanti-inflammatory drug whose sale amount is the most. It has theR-enantiomer and S-enantiomer, which the main pharmacodynamic action isreduced by the S-ibuprofen while R-ibuprofen results in adverse effects. It canreduce dosage, improve treatment effect and reduce adverse effects if we canobtain S-ibuprofen by the chiral resolution. Nowdays, the chiral resolution islimited because high cost and tedious steps. We can obtain S-enantiomers bychiral liquid membrane which is used L-tartrate as chiral resolution regent. Itis simple to operate and reduce the cost of chiral resolution.A HPLC method for ibuprofen enantiomers in the aqueous of chiralextraction and chiral liquid membrane was established. An AGP column wasused with sodium phosphate buffer solution consisting methanol as organicaddictive regent, in which the pH was7.0. The detection wavelength was setat220nm and the flow rate was0.5mL/min. The method could improve thebaseline shifting and column pressure increasing and could analyse theconcentration rapidly and effectively. It also reduced analytical cost andimproved the lifetime of the chiral columu. L-pentyl tartrate was the best chiral extraction for racemic ibuprofen andthe optimum concentration was0.2mol/L. The distribution and coefficientand separation factor were much better when polar organic solvent was usedand reduced with the increase of the pH in the aqeous phase.Racemic ibuprofen was successfully separated by chiral liquid membranewhen L-pentyl tartrate solved in octane was used chiral membrane phase inthis paper. The best pH in the stipping side was2.5and the separation factorwas about1.2. The ee value reached16%by cascade experiment. Theinfluence of operation conditions on the resolution were studied.Mathematical models of optical purity of ibuprofen in the stipping phase withthe NTU was established. The applicability and accuracy of the model wereverified by some data in the cascade. The chiral liquid membrane based onL-tartrate reduced the cost. It was of significance to promote theindustrialization process of chiral drugs. |
