| L.macranthoides Hand.-Mazz known as dajingyinghua, belonging to agiosperms, eudicots, core eudicots, dipsacales, caprifoliaceae, lonicera, distributed mainly in Anhui, Jiangxi, Hubei, Hunan, Guangdong, Guangxi, Sichuan and Guizhou provinces.L.macranthoidesHand.-Mazz.is one of the bulk medicinal herbs by artificial cultivation in our province and the main products of Lonicera Japonica in our province. It is widely planted in Longhui, Xupu, Xinshao, Dongkou, Xingning, Linwu, Anhua, Leiyang, Guiyang, etc, accounting for more than90%of Lonicera Japonica in our province, of which Xupu and Long hui’s yield is the highest, so they are the standardized production base counties of Chinese herbal medicine in hunan province. In the year2001Longhui was awarded as "the town of chinese honeysuckle" by State Forestry Department, and Xiaoshajiang town becomes the biggest collecting and distributing center of honeysuckle in China.With the function of clearing heat, detoxification, inhibition, anti-inflammation, and antioxidation,ect, L.macranthoides Hand.-Mazz is the dried flower bud or the initial flower (Flos Lonicerae Confusae is called by Pharmacopoeia,Lonicera Japonica is called by folk)is widely used in drinks, medicine and daily living supplies. In this thesis the Extraction technology, Determination of the content and difference, Purification technology were mainly studied on macranthoidin B and dipsacoside B in L.macranthoides Hand.-Mazz, the results are as follows:1.Comparison of three kinds of extraction method:warm maceration extraction method, ultrasound extraction method and hot reflux extraction method. The results show that:hot reflux extraction method and ultrasound extraction method is superior to warm maceration extraction method, and hot reflux extraction method is slightly better than ultrasound extraction method. Due to the hot reflux extraction method is comparatively mild, the degree of macranthoidin B degradation is small, and mass production is easier, Therefore the experiment take hot reflux extraction method.2.Comparison of four kinds of solvent:water, methanol, ethanol and ethyl acetate on the two kinds of saponins extraction effect. The results show that:the polarity of the two kinds of saponins is relatively large, easily soluble in methanol, ethanol and other alcohols, difficult to soluble in ethyl acetate and low polarity solvent. Considering the methanol is toxic, therefore using ethanol as extractant.3.Groping for alcohol hot reflux extraction method on the two kinds of saponins extraction effect. Through single factor experiment building extraction process, and on this basis take four factors and three levels orthogonal experiment. The optimum extraction process parameters as follows:extraction temperature is80℃extraction time is120min, ratio of material to liquid is1:10,ethanol concentration is90%.Under the optimized conditions for validation experiments, its total content reached7.18%. Indicate that hunan L.macranthoides Hand.-Mazz in these2kinds of saponin content is much larger than the Chinese pharmacopoeia (2010) prescribed in Flos Lonicerae Lonicera macranthoidin B and dipsacoside B content shall not be less than5.0%.4.Establishment of the HPLC determination method of macranthoidin B and dipsacoside B. The chromatographic conditions as follows:Chromatographic column is Hypersil ODS (4.6×250mm,5μm)detection wavelength at210nm, column temperature at40℃,Chromatographic velocity at0.6mL/min,With acetonitrile and0.4%acetic acid for gradient elution. Under the chromatographic conditions chromatographic peak is complete,degree of separation is fine, showed that HPLC method detection method is reliable, feasible.5.Analysis of different processing methods, different medicinal parts and different varieties of total content of macranthoidin B and dipsacoside B in L.macranthoides Hand-Mazz. The results show that:hunan L.macranthoides Hand.-Mazz in different varieties, the two kinds of saponin content are all in7%, the content difference is small. Compared sun drying, bake drying and vacuum freeze drying method, sun drying method cause a great impact on the two kinds of saponin content, the two kinds of saponin content is only6.5%.The root of the two kinds of saponin content is about1%,far less than the content in flower bud.6.Study of SA-1type medicinal saponin adsorption resin on the two kinds of saponin purification process. Observe the factor of sample concentration, ph value.flow rate on the adsorption rate influence and elution concentration, flow rate on the desorption rate influence. The results showed that:flow rate was2BV/h, sample concentration is about2.5mg/mL, pH=6.8and at the speed of2BV/h, ethanol concentration of60%as the elution conditions, obtained the purity of macranthoidin B and dipsacoside B was65.6%. |
