As a carotenoid compounds, fucoxanthin has many biological activities, itsdistribution is also very extensive. We used the ultrasonic composite enzymaticmethod to extract fucoxanthin from kelp in this paper. We mainly studied the optimalprocess parameters of extracting fucoxanthin, saponification process and the stabilityand oxidation resistance of fucoxanthin. We discussed various factors on fucoxanthinextraction rate during the extracting process, determining the optimal conditionthrough orthogonal test. We analyzed of the single factor influence during thesaponification process, determining the optimum parameters through orthogonal test.This paper also studied the effect of the outside factors on stability and theantioxidation in vitro.The main research contents and results are as follows:1、The topic mainly studied the optimal parameters of ultrasonic extraction,analyzing the extraction time,ratio of material to liquid, temperature and other factorson fucoxanthin extraction rate. The experimental results showed that, by adding1%ascorbic acid, with1:45ratio of solid to liquid adding anhydrous ethanol, the initialtemperature of60℃, ultrasonic treatment30min, extracting for2times.In the end,the fucoxanthin extraction rate is0.4417mg/g.2、The topic mainly studied the optimal parameters of enzymatic extraction,analyzing the enzyme concentration, extraction time, extraction temperature and pHon fucoxanthin extraction rate influence. The experimental results showed that, byadding7.5%of cellulose and2.5%pectinase, pH4, at40℃for37min. In the end, thefucoxanthin extraction rate is1.0068mg/g.3、The topic mainly studied the optimal parameters of saponification process,analyzing the content of KOH, saponification time and saponification temperature onfucoxanthin saponification rate influence. The experimental results showed that, byadding20%of KOH, at30℃for1.5h. In the end, the saponification rate is1.3540.Through the process of extraction to remove the KOH and silica gel columnchromatography for further separation and purification. In the end, the fucoxanthinextraction rate is0.9012mg/g.4、The topic studied the influence of factors such as light, temperature, ascorbicacid, reducing agent, acid, alkali on stabilitu, the results confirmed fucoxanthin instability. The topic studied the scavenging ability of fucoxanthin on hydrogenperoxide, hydroxyl radicals (OH) and DPPH free radical, which confirmed theantioxidation of fucoxanthin, laying the foundation for further research.
