This article mainly studies the chemical property of colchicine extracted from lily, and its interaction with BSA and DNA by electrochemical method. A new glassy carbon electrode midified by phenylenediamine and multi-walled carbon nanotubes on its surface is made after research of modified electrode. Besides, a binding number and binding constant determination method of colchicine with BSA or DNA was also put forward. The method achieves a result close to the results of spectrophotometry and advantages in method difficulity, speed and cost.1. Studies on colchicine is always a hot research topic because of its great pharmaceutical value. Electrochemical method is a preferred method of determing colchicine as it’s easily operated and micro sample consumption. We seek matariels more suitable for midifying working electrode, and choose phenylenediamine and multi-walled carbon nanotubes for the first time in this field. The characteritics of modified electrode is studied by a differential pulse voltammetry method.2. Interaction between colchicine and BSA is studied by el eclectro-chemical method. Colchicine and BSA combine and form super molecules which cause decrease of oxidation peak current. Binding number and binding constant are calculated according to experimental data. The article also probe into change of binding capacity between colchicine and BSA in different mediums, which greatly affects the binding capacity. The decrease of oxidation peak current has a linear relationship with concentration of BSA. The linear range is1.5×10-7~2×10-6mol/L, and the detection limit is4.0×10-8mol/L.3. Interaction between colchicine and DNA is also studied, and find that single-stranded DNA differs from double-stranded DNA in mechanisms of interaction with colchicine and binding capacity. A simple and feasible electrochemical method with a high sensitivity is studied in this article, and we get the binding constant without expensive equipment. Besides, UV spectrophotometry metohd is also introduced to study the binding mechanism.
