Research on colchicine at domestic and abroad was reviewed, including resources of colchicine, methods of extraction, methods of separation and characterization.The extraction and separation conditions of colchicine from lilium were studied. The results showed that colchicine was obtained from lilium by alcohol with a ratio of 1:3 (material : solvent) at room temperature for 12 hours. The extraction rate of colchicine was 91.37%.A column chromatography was made for separation of colchicine from lilium. The stationary phase was Al2O3, And eluting agents were CH6Cl3 and CHsOH with a ratio of 20:80. The amount of sample was 1/30 the amount of Al2O3. The resultant could be obtained from eluting liquid concentrated.Colchicine had been measured with scan polarography in a 0.6%(C4H9 )4NCl-B.R.buffer solution (pH1.81). An excellent linear relationship existed between the peak current and the concentration of colchicine (2 X 10~7mol/L~4X10~5 mol/L). The detection limit is 4.0X10~8mol/L, and the relative standard deviation was 2.0%. The recovery was 98.0%~106.0% . This method was successfully applied to determining colchicine in tablet of colchicine and lilium.Chemical components of lilium were researched with volumetric method, spectrophotometric method, atomic absorption spectroscopy and gravity method. The results showed that reducing sugar 2.11%, sucrose 1.82%, protein 4.83%, vitamin B,0.036%, vitamin B20.22%, vitamin CO.0161%, pecton 1.619%, Fe 0.0024%, Mg 0.025%, K 0.4211%, and alkaloid 4.2906mmol/100g.
