Abstract: Quinolones is a synthetic antibacterial drugs, because of its strongantibacterial activity, broad spectrum antimicrobial, pharmacokinetic characteristics,it is widely used in the prevention and control of animal diseases, especially itsthird-generation quinolones-fluoroquinolones. Because the drug abuse and not strictimplementation of the withdrawal period system, it cause excessive residue in animalfoods, long-term consumption of such foods will cause a series of different levels ofside effects, therefore, residue testing of the drugs in animal foods is very important.This study selected chicken liver and pork liver as raw materials, the sample wasseparated by solid phase extraction(SPE)cartridge and determined by Flow-injectionChemiluminescence, results were as follows:1. The method was based on the chemiluminescence(CL) reaction betweenluminol and potassium periodate was sensitized by danofloxacin mesylate in alkalinesolution, the sample was separated by solid phase extraction (SPE)cartridge, wasdetermined by Flow-injection Chemiluminescence under the same conditions with thestandard curve. Under optimum conditions, sodium hydroxide concentration was0.40mol/L, luminol concentration was1.0×10-3mol/L, potassium periodateconcentration was1.0×10-4mol/L, the good linear relationship between thechemiluminescence intensity and the negative logarithm of danofloxacin mesylateconcentration was kept in the range of4.53×10-62.27×10-3mg/mL. The detectionlimit of danofloxacin mesylate was3.76×10-6mg/mL, the relative standard deviationRSD for the determination was1.9%(n=11), the recovery rate with added specimenwas held in the range of69.00%-97.50%.2. It was found that in alkaline solution, the chemiluminescence produced by thereaction between luminol and hydrogen peroxide was inhibited by difloxacinhydrochloride, the sample was handled by solid phase extraction (SPE) cartridge anddetermined with this system, a new method of Flow-injection Chemiluminescencedetermination of difloxacin hydrochloride in the pork liver and chicken liver wasproposed. The results showed that sodium hydroxide concentration was0.10mol/L, luminol concentration was7.0×10-4mol/L, hydrogen peroxide concentration was0.20mol/L, the good linear relationship between the chemiluminescence intensity andthe negative logarithm of difloxacin hydrochloride concentration was kept in therange of1.00×10-91.00×10-5mg/mL,the detection limit of difloxacin hydrochloridewas4.07×10-9mg/mL, the relative standard deviation RSD for the determination was0.93%(n=11), the recovery rate with added specimen was held in the range of52.50%-78.63%.To establish a trace analysis method of Flow-injection Chemiluminescencedetermination of danofloxacin mesylate and difloxacin hydrochloride in animal liver,provide a simple and rapid detection of new technologies, ensure the safety of animalproducts in order to better guarantee the people ’s food security.
