China’s kelp production is currently ranked frist in the world and the output of thekelp industrial have accounted for more than60percent of world production, whichcontains a large of kelp residue. There is a large number of cellulose resources in kelpresidue.In this thesis the high quality pharmaceutical excipient microcrystalline cellulosewhich meet the pharmaceutical standards was made, which not only provide a reliabletheoretical basis for the use of the waste in seaweed chemical industry but also expand thetypes of microcrystalline cellulose to provided a new choice for pharmaceutical excipient.This project were carried out in four parts In the first part, the extraction of cellulosein kelp residue was determined, the content of cellulose is91.38%, the ash content is3.02%,water holding capacity is2650.33%,and swelling force is34.23mL/g of cellulosein kelp residue are better than the wheat bran cellulose which was used as the referencesubstance(the content of cellulose is89.38%, the ash content is2.76%,water holdingcapacity is678.67%,and swelling force is7.67mL/g).The kelp cellulose which made in the first part were used as the raw material in thesecond part. The acid hydrolysis process, the drying process and the process of grindingwere studied, the optimum conditions for the preparation of microcrystalline cellulosewere investigated. The best process as follow: concentration of hydrochloric acid:2mol/L;the ratio of kelp residue fiber and hydrochloric:1:10;the hydrolysis temperature:100℃;hydrolysis time:40min, The method of vacuum freeze-drying milling crushed can be used.The index of MCC which made under the above conditions are listed that: the content ofMCC is that98.23%;the polyer:138.3; whiteness:89.1%, the moisture content:2.25%.The cellulose in kelp residue which made by the optimum process can meet the standard of ChP (the edition of2010).In the third part the characterization of homemade kelp microcrystalline cellulosewere studied by the method of X-ray diffraction and SEM, which used the commercialsamples as reference. The results showed that the homemade seaweed celluloses’ physicaland chemical indicators in line with the pharmaceutical excipients requirements. Thefineness, pH, the substance dissolved in the water, chloride, starch, loss on drying, residueon ignition, heavy metals, arsenic salt in homemade kelp microcrystalline cellulose wereinspected in accordance with the pharmacopoeia, the result can meet the standard of ChP(the edition of2010).In the fourth part, the pharmacy parameters which including fluidity, compressibility,swelling of self-made kelp microcrystalline were determined which used thecommercially available microcrystalline cellulose as the standard substance. Theparameters which including tablet hardness, disintegration, dissloution of Aspirin-baseddrug tablets which made by the method of dry granulation were studied. The table agentwere developed using the self-made microcrystalline cellulose. The study of cellulose as ahealth food was carried out. Comparison of wheat bran dietary fiber table agent on therole of the patients with constipation was made. The result showed as follow: theself-made kelp microcrystalline cellulose’s fluidity is better than the commerciallyavailable microcrystalline cellulose. Aspirin tablets’ disintegration, dissolution is alsosuperior to the commercially available microcrystalline cellulose and the kelp slagmicrocrystalline cellulose table agent has more superior help defecate runchang efficacy.
