The present study was undertaken to optimize the extraction technology of Caulis Lonicerae and Sophorae tonkinesis and Licorice from qingyan granules and to engage qualitative and quantitative analysis for these medicinal materials. Two active components of each herb were selected as the standard. The qualitative analysis combined thin-layer chromatography and high-performance liquid chromatography and the quantitative analysis by reversed-phase high-performance liquid chromatography was performed to establish the quality standards for the qingyan granules. The extraction technology of water extract-alcohol extraction, ultrasonic extraction and microwave extraction was optimized by L9(34) orthogonality experiment. By comparison, the best proposal for the extraction of qingyan granules was finally established.The six active ingredients of qingyan granules, including chlorgenic acid, caffeic acid, matrine, oxy-matrine, liquiritin and glycyrrhizic acid, were analyzed by the qualitative analysis with thin-layer chromatography and high-performance liquid chromatography. These above-mentioned active ingredients was found that were all existed in the qingyan granules. Moreover, microwave extraction caused no structural damage or degradation of them. The quantitative analysis for these active ingredients was established by reversed-phase high-performance liquid chromatography, with each chromatographic retention time less than15minutes. The peak shape was found to be clear and symmetry, which revealed that the active ingredients could be completely separated from the endogenous impurities of qingyan granules.The extraction technology for the above three medicinal materials was optimized by the univariate analysis and L9(34) orthogonality experiment. By comparing the methods of water extract-alcohol extraction, ultrasonic extraction and microwave extraction, the major effect factors in orthogonality experiment were well defined. The present study showed that the extraction percentage of microwave extraction was similar to water extract-alcohol extraction and ultrasonic extraction, while its extraction time was faster and the operation was simpler. Thus, the microwave extraction was suitable for production in large scale and also can save the energy greatly. The best proposal for the extraction of qingyan granules was3times of extracting under700W by microwave extraction, with14minutes per time.
