| According to the reports of "Fu" brick tea process and health care function, Eurotium Cristatum which is also called the "golden-flower" fungus on "Fu" brick tea was isolated and purified. The optimal condition for culturing the "golden-flower" fungus was discussed. While commercial green tea was chosen as row material, the fermented loose tea was made by artificial inoculation. Then changes of chemical components and testing were carried out during the fermentation process. To discuss the possibility for producing a new type of dark tea by artificial inoculation, the anti-obesity effect of the dark loose tea on rats was analyzed. The results were as fallows:1. The optimization culture of the "golden-flower" fungus is:at30℃in6%sucrose and0.15%ammonium dihydrogen phosphateis medium for culturing96h, with a shaking speed of120r/min. This liquid culture suspension was prepared for the subsequent inoculation.2. The fermentation process:The fermentation process of pan-fired green tea contained five steps:pretreatment of green tea (screening, sorting tick, moisturizing with5%water); packing(100gram per bag); sterilization on microwave and cooling(400W,90s);15%Eurotium Cristatum was added in green tea and artificial inoculating, fermented for7days at30℃and dried at60℃. At last, the dark loose tea had golden appearance, pure aroma with aromatic flavor, mellow and mitigated bitter taste, which accorded with the characteristic of traditional "Fu" brick tea.3. The radical scavenging tests:The scavenging abilities of the dark loose tea were reached75%and56%respectively in determination by (1,1-Diphenyl-2-icrylhydrazyl) and ABTS (2,2’-Azinobis-(3-ethylbenzthiazoline-6-sulphonate) radical methods, and were similar to those of "Fu" brick tea produced naturally from Hunan province, and were higher than those of "Fu" brick tea produced naturally from Zhejiang province.Though the fungus in dark loose tea was isolated from natural "Fu" brick tea produced in Zhejiang province, the fungus grew better and the radical scavenging abilities of dark loose tea improved after the cultivation medium optimization.4. Lipid-lowering effect:the anti-obesity function of natural and artificial fermented "Fu" brick tea was discussed. The results indicated that two kinds of "Fu" brick tea can both inhibit the gain in weight, the indexes including body weight, fat wet weight, fat index, TG and TC are all lower than the obesity model group, while HDL-C improve significantly, the HDL-C contents of natural and fermented "Fu" brick tea were higher than control40%and17%respectively. The AIP levels of green tea, natural "Fu" brick tea and dark loose tea rats obviously decreased53%,55%and67%respectively than those of fat rats group, especially, the AIP level in dark loose tea rats was close to that in normal group rats. |
