This article aim at low utilization oil rate, almond meal can not be comprehensive utilization. The study focuses on the development and utilization of aqueous enzymatic method in sweet almond. This article studied on sweet almond oil extraction rate of aqueous enzymatic method and its protein, getting the best extraction process parameters, simultaneously in order to realize the extraction of sweet almond oil and protein, the oil and protein quality were studied also. For enterprises to extend the industrial chain, to develop new products and improve the output value of enterprises laid a theoretical basis. The results are as follows:(1) This paper studied the influence of pretreatment conditions on the sweet almond oil extraction rate, based on the single factor experiment, used the Design-Expert7.1.6Trial software designed response surface optimization experiment. The results showed that the pretreatment process can obviously improve the sweet almond oil extraction rate, the optimum process parameter is the temperature at52.7℃, the heating treatment time at18min, the breaking time at7min, the enzymatic extraction ratio of sweet almond oil was up to55.44%.(2) Researched one step enzyme and two steps enzyme. The enzymatic hydrolysis extraction rates were:51.11%and56.31%. Finally, choosed the one step method.Based on the basis of single factor experiment,conduct response surface, the optimized process parameters for the best:enzymolysis temperature of57.81℃, enzyme amount to3.46%, solid-liquid ratio to1:6, enzymolysis time4.48h, by optimizing the sweet almond oil extraction rate was up to60.80%.(3) Researched the demulsification process, the effect on different centrifugal conditions of demulsification, on the basis of single factor experiment and orthogonal test, finally obtains the influence order of various factors on sweet almond oil extraction rate: centrifugal speed> centrifugal time> pH value, the best demulsification process parameters were:centrifugal speed was12000r/min, centrifugal time at21min, the pH value was4.0, the optimized oil rate was75.76%.(4) Using GC-MS qualitatived on the enzymatic hydrolysis of sweet almond oil, refined oil and commercial oil fatty acids, quantitative analysis of available were:the unsaturated fatty acid content in enzymatic hydrolysis on sweet almond oil and refined oil were:95.16%and93.50%; the unsaturated fatty acid content in New Food sweet almond refined oil is:94.86%; at the same time analyzed on the physical and chemical indexes of almond oil.(5) Through laboratory simulation plant press production process condition for the production of sweet almond oil, sweet almond oil yield of61.8%. According to the actual situation of factory equipment, in the factory simulation laboratory under conditions of enzymatic extraction technology of sweet almond oil, sweet almond oil yield of52.64%. Optimization the enzymatic hydrolysis technology, the finally enzymatic extraction rate was75.76%.(6) According to the standards prescribed by the state, by amino acid analyzer and Automatic Kjeldahl nitrogen apparatus analysised of the physical and chemical properties of almond meal, the content of protein24.4%, fat9.25%and the color of the enzyme protein is milky white.
