In this thesis, the synthesis of5-bromo-4-substituted pyrimidine derivatives1-22was studied by the Grignard reagent addition method.First,5-bromo-4-substituted-3,4-dihydropyrimidyl intermediates were synthes-ized by the nucleophilic addition reaction of Grignard reagent to5-bromopryrimidine.2-Methyltetrahydrofuran was investigated as the best solvent, and the reaction time was optimized.5-bromo-4-substituted-3,4-dihydropyrimidyl intermediate was futher oxidized to form5-bromo-4-substituted pyrimidine derivatives. It was found that when KMnO4used in acidic system at0℃, twenty products were obtained in10%to70%yields. When33%KOH and K3Fe(CN)6were used in alkaline system at room temperature, twenty-one products were obtained in15%to70%yields.The structures of all the products were characterized though1H NMR and13C NMR. |
