This paper summarizes the theory and application scope of the evolving factor analysis and alternating least squares in multivariate resolution methods. In addition, these algorithms combining with spectrophotometry were applied to identify overlapping peaks in complex system, the pure spectral curve and concentration changing curve of one-component in system were resolved, and the qualitative and quantitative analysis of the components were carried out.1、Definition and development of Chemometrics and multivariate resolution methods is introduced. Several related multivariate resolution methods were introduced simply, which focuses on presenting the theory of evolving factor analysis and alternating least squares and the application condition of these two algorithms. The application of multivariate resolution methods in determining the number of components and the resolution of overlapping peaks were also described in detail.2、The nature of the curcumin and its degradation process were studied through the multivariate resolution alternating least squares (MCR-ALS) combining with spectrophotometry.(1) The absorption spectrum of curcumin in different pH conditions were analyzed by MCR-ALS to obtain the one-component spectral curve and concentration changing curve existing in system, and the form of curcumin in different pH conditions were resolved.(2) The absorption spectrum of the degradation of curcumin were analyzed by MCR-ALS, the results showed that uv-vis spectral curves of curcumin were similar in acidic, neutral and alkaline conditions, and the dynamic curve of the degradation process of curcumin were also analyzed.3、The reaction mechanism of Homoharringtonine with calf thymus DNA were investigated by MCR-ALS combing uv-vis spectrophotometry, fluorescence spectrophotometry. Augmented column and row-wise data matrix were analysed by MCR-ALS to determine the number of component in the reaction system, and to resolve the pure spectral curve and the concentration curve of each component. The binding constant of reaction K1can be calculated from the analytical curve of concentration distribution,.K1=3.2×105M-1. It is deduced that the action mode of Homoharringtonine with DNA is intercalative binding by cyclic voltammetrys and viscometry.4、 The reaction mechanism of derivative isosteviol with calf thymus DNA were investigated by MCR-ALS combing uv-vis spectrophotometry, fluorescence spectrophotometry. As the spectral overlap is serious in system, augmented data matrix was analysed by MCR-ALS to determine the number of component in the reaction system, as. well as the pure spectral curve and the concentration curve of each component. The result indicated that the augmented data matrix can reduce rotational ambiguities and resolve the problem of rank deficiency. The intercalative binding mode of derivative isosteviol with DNA was deduced by viscometry.
