Vanadium has been proposed to act an important role in the glucose metabolism and to be an essential trace element in human body. Vanadium compounds can simulate the physiological effects of insulin. So far, a large number of study show that some inorganic vanadium compounds have well insulin effects, though inorganic vanadates has lots of drawbacks, for example:toxic to humans, bad fat-solubility in humans body, big dose of taken in therapgy. So, reseachers always select organic ligands and coordinated atoms to improve these problems. Currently, organic vanadium compounds have been the popular topics in bio-inorganic chemistry.We designed two series of vanadium complexes in this thesis, including: vanadium-hydro xylamido complexes and Schiff base compounds, both series consist of N\O-containing ligands. The reachers had studied the protein tyrosinase activity-PTP1B activity. More details as follow:(1). Six unknown vanadium-hydro xylamido complexes with L-Proline, L-Hydroxyproline, L-Tyrosine, Pyrazole-3-carboxylic acid,2-Piperidinecarboxylic acid,3-Amino-2-pyrazinecarboxylic acid has been made. In addition, we synthesised of two known complexes. Among that, the structure of the first two compounds have been determined by the single crystal diffraction analysis.(2). Four new Schiff base ligands and two new vanadium complexes with Schiff base ligands were synthesized in the ethanol solution. The Schiff base involve salicylaldehyde-2-amino-5-hydroxy benzoic acid, salicylaldehyde-2-amino-5-methyl benzoate, salicylaldehyde-2-amino-5-chlorobenzoate, salicylaldehyde-2-amino-6-chlorobenzoate. After that, we used several analytical methods to study the composition of the complexes.(3). To understand the performance of our new compounds, several tests were taken, such as:determination of the partition coefficient (fat-solubility reserch), activity test, (PTP1B the protein tyrosine phosphatase experiments). The preliminary conclusion is a basis for further research and establishment of structure-activity relationship of vanadium complexes as insulin mimics.
