The Research Of Microorganism Aerobic Metabolic Activity Fast Detector

It is an important method of using microbial to degrade the environmentalpollutants in bioremediation. On environmental microbiology study, we often need todetect the microbial metabolic activity which was separated from environmental for theidentification of microorganisms. At present, the research on aerobic microbial metabolicactivity is very tedious, and the analysis of the test procedure is complicated and theequipment is expensive, the cost is very high, so we need to set up a convenient and quick,cheap method.In this paper, we get three bacteria strains of A₃, B₄, M₁which are from thewastewater. The identification is carried out from observing the outline and structure, thephysiological and biochemical tests and16S rDNA analysis. The results showed that thethree bacteria lake respectively Solutions bacillus, Bacillus licheniformis and Brewersyeast.Using the three bacteria as molecular recognition elements, they are immobilizedby sandwich with the use of cellulose porous membrane of0.45μm. The biosensor iscomposed by connecting the immobilized microorganisms membrane and thepolarographic oxygen electrode. The response properties and optimum measurement fortoxicity microbial sensor were investigated. The experiment indicated that, the optimaltemperature is30℃, the appropriate pH range is6.5～7.0.Using microorganism aerobic metabolic activity fast detector to measure themetabolic activity of7kinds of substances, and compared with Biolog method. Resultsshow that this project instrument developed by the measured results have consistency withBiolog methods, the instrument has the certain application value.
