Research In Chinese Herbal Drugs Against NDV And Animal Experiment Of Compound Recipe

【Objective】In order to triturate the compound recipe against NDV,25kinds of chinese herbal drugs and thebest compound recipe were chose by cell culture model in vitro,the clinical potency of compound recipe wereestimated by animal models which infected Newcastle disease,also safety of compound recipe wasestimated.By aforementioned work,it will supply theoretical and practical evidence for researching newantiviral drugs.【Method】(1)In BHK-21、HeLa and DF-1cell lines,the susceptivity of NDV was estimated by Cytopatho-genic Effect(CPE),hemagglutination test(HA) and molecular biological method,the optimal cell line wasdetermined to proliferate for NDV.(2)The effective components were extracted from Chinese herbal drugs bywater decoction,and the antiviral potency were evaluated by different ways of direct action,prevention andtreatment,the potency of anti-NDV of different drugs were determined by therapeutic index(TI).(3)Fourcompound recipes were composed according to traditional chinese medicine theory,and the best matching ofcompound recipe was determined by above ways.(4)The drug dose were determined by toxicological test,andthe median lethal dose(LD50) was determined by chicken which artificially infected NDV,then the curativeeffect of compound recipe was determined by preventive and therapeutic test on chicken.【Results】(1)The result showed that NDV produced typical CPE with cytomixis and syncytia after serialpassages in DF-1and BHK-21cell lines,and the CPE characterized by rounding,degeneration in HeLa cellline.But the susceptivity of NDV and HA titer were better in DF-1cell line than others,the HA titer was1:16passaged in fourth generation in DF-1.By molecular biology method,the NDV was identified as a virulentstrain.(2)The direct action showed that Radix Astragali Mongolici,Flos Dendranthematis Indici,RadixPulsatillae,Herba Verbenae Offcinalis, Herba Asari Mandshurici,Flos Lonicerae,Radix Scutellariae Baicale-nsis,Herba Agastaches Rugosae and Radix Sanguisorbae were effective,the TI were64、16、32、32、64、64、32、32、64respective.The prevention indicated that Radix Astragali Mongolici,Radix Pulsatillae,FlosLonicerae,Flos Dendranthematis Indici,Herba Asari Mandshurici,Radix Sanguisorbae and Radix BupleuriChinensis were effective,the TI were16、64、64、16、32、16、16.And the treatment indicated that RadixAstragali Mongolici,Flos Lonicerae,Flos Dendranthematis Indici,Herba Verbenae Offcinalis,RhizomaChanxiong,Radix Bupleuri Chinensis,Herba Agastaches Rugosae and Herba Asari Mandshurici couldrestrained replication of NDV,the TI were32、16、16、16、16、8、8、8.In above three ways,it indicated thatRadix Astragali Mongolici,Radix Bupleuri Chinensis,Radix Pulsatillae,Flos Dendranthematis Indici,FlosLonicerae,Radix Scutellariae Baicalenhsis,Herba Agastaches Rugosae,Herba Asari Mandshurici,RadixSanguisorbae,Rhizoma Chanxiong and Herba Verbenae Offcinalis were strong effective against NDV.(3)As awhole,seven anti-NDVdrugs which were Radix Astragali Mongolici,Flos Dendranthematis Indici,RadixBupleuri Chinensis,Radix Scutellariae Baicalensis,Flos Lonicerae,Radix Sanguisorbae and Herba AgastachesRugosae were chose,so four compound recipes were organized,finally determined the best compound recipewhich was made of Radix Astragali Mongolici,Rhizoma Atractylodis Macrocephalae,and so on.The TI ofdirect action,prevention and treatment were16、64、32respective,and the protective rate were34.29%、37.86%、56.71%.(4)The drug dose of compound recipe was0.5~2g/mL,and the LD50was10-2.76in vivo.Thepreventive and therapeutic effect of compound recipe indicated that it could decline the mortality,but theprevention was better than treatment,the protective rate of high-dose、middle-dose and low-dose group were70%、40%、10%,while the protective rate of therapeutic effect were30%、20%、20%,the protective rate ofribavirin group were10%、30%respective.【Conclusion】(1)DF-1cell line was determined the optimal cell to proliferate for NDV.(2)Eleven kinds of anti-NDV drugs of Radix Astragali Mongolici,Radix Bupleuri Chinensis,Radix Pulsatillae,Flos Dendranthem-atis Indici,Flos Lonicerae,Radix Scutellariae Baicalenhsis,Herba Agastaches Rugosae,Herba Asari Mandshu-rici, Radix Sanguisorbae,Rhizoma Chanxiong and Herba Verbenae Offcinalis were chose by three ways ofdirect action,prevention and treatment.(3)The best compound recipe which was made of Radix AstragaliMongolici,Rhizoma Atractylodis Macrocephalae was chose.(4)The preventive and therapeutic test wereconfirmed in chicken which artificially infected NDV,it showed that the compound recipe could resistinfection of NDV, and decline the mortality of chicken after infected NDV,also lengthen chicken′survivaltime.
