Alien addition lines are importent materials for gene assignment and crop genetic improvement. In order to generate the disomic alien addition lines and substitution lines in Brassica vegetables, the Chinese kale (Brassica alboglabra Bailey)-Flowering Chinese cabbage (Brassica campestris L. var. utilis Tsen et Lee) monosomic alien addition lines were used as materials, and by means of artificial hybridization, chromosome number examination, karyotype analysis, morphological observation and SSR identification, the transmission of alien chromosomes were determined and disomic alien addition lines and substitution lines were generated. The main results are showed as follows:1. The chromosome number examination for the progenies from the reciprocal cross between the Chinese kale - flowering Chinese cabbage monosomic addition lines and recipient parent showed that the transmission rates of n+1 gametes ranged from11.46% to 37.35% for female and from 3.70% to 12.90% for male.2. Correlation analysis showed there were some correlativity between the transmission rates of n+1 gamete and the rates of trivalent formation at diakinesis, the rates of chromosome distribution with 9/10 at first anaphase, the plollen viability, the pollen amount and chromosome relative lentgh, but no one reached the significance level. The correlation degree was chromosome distribution, trivalent, viability, chrosome relative length and plollen amount, respectivelly.3. Via karyotype analysis, meiotic observation, SSR identification and morphological comparison, five disomic alien addition lines CC+2A2、CC+2A3、CC+2A6、CC+2A8 and CC+2A9 were obtained from the selfed progenies of the Chinese kale– flowering Chinese cabbage monosomic alien addition lines. There are no obvious differences between the monosomic and disomic addition lines in main phenotype traits. However, the fertility for the disomic alien addition lines is much better than those of the monosomic addition lines.4. Based on the partial homology between A and C genome in Brassica, using the monosomic alien addition line CC+A9 which has a higher trivalent formation as parent, a substitution line was obtained from its selfed population by cytological, morpholocial and SSR identification, which is the first substitution line in Brassica plants so far. 5. A primary trisomic line of Chinese kale was obtained from the progeny of No.10 monosomic alien addition line.
