Hybrid identification was studied in cross-breeding of cold-resisting Mei flower. The hybrids (4-8a) were analysed by morphological detection and molecular marker analysis, promoting selection of progenies and developing applicability of molecular maker determination in hybridization of Mei flower. The main results obtained are as follows:1. SSR molecular maker was used in parentage determination of hybrids by Mei flower and closely related species.47SSR primers from Mei flower’s closely related species and3EST-SSR primers were employed to amplify25parents. Specific bands were detected in22male parents by18primers, with a high producing rate of95.7%.69hybrids belonging to13cross combinations were analysed, of which41progenies were identified to be authentic using10of the18primers. The discrimination rate was62.1%. The result indicated a certain feasibility of SSR primers from closely related species in Mei flower’s hybrid detection, which was an innovation in certain field.2. AFLP molecular maker analysis was applied to detecte the parentage among92Mei flower hybrids and28suspected parents. According to the DNA fingerprints by8primer pairs and the clustering analysis,’Dan Fenghou’was the possible female parent of most of the F1crossbreds and the male parents belonged to either true Mei cultivars or closely related species. The rest hybrids were failed to detect the possible parents as no specific bands were found.3. comparing the F1crossbreds with parents by13blooming phenotype traits and7vegetation traits, finding that all the19hybrids were authentic; and the other92progenies were assumed potential cross-combinations on the basis of morphological correlation and AFLP analysis.
