| Lychnis fulgens Fisch. is a herbaceos perennial in Caryophyllaceae. Oragen flowers born in the top of plant which give it a high ornamental value. It can be used as garden plant as well as ground cover plant. It also has been chosen as the Beijing Olympic landscape display flowers. It is a wild native flower with great market potential for development. On the basis of the studying in the process of meiosis in pollen mother cell(PMC) and megaspore mother cell(MMC), we carried out the experiment colchicine induction of polyploid in Lychnis fulgens using different methods. On discovering a high germination rate method, we studied induction of polyploidy by treating the germinative seeds. Establishment of aseptic system give the platform to carry out expriment of colchicine soaking with the stems to induced polyploid. The main experimental results can be summarized as follows:The expriment of observation on the meiosis of PMC and MMC shows that there is a relationship between the meiosis stages and its morphological characteristics. When the flower bud was14mm in length, most of microspores developed into mononuclear aside period,that of megaspore mother cells just entered pachytene.After injection treatment of colchicine, the hurting part would turn black and cause the failure of blossom.Seed setting rate was0.Seeds and seedlings were obtained from the cotton dip treatment, but seed set rate and seedling rate was far lower than the control. With the increasing concentration, seed rate and seedling rate reduced. One petal variation plant was obtained from the treatment of0.1%concentration of colchicine and36duration.After high temperature treatment, the plant can blossom but the seed set turn out to be0.Using150mg/l of GA3pretreated the stored seeds for24h sown in petri dishes can get the highest germinative force of76.33%and the highest germination rate of85.33%.Variative plants were obtained under each colchicine concentration and concentration of0.10%was the optimal induction concentration with the higher mutagenic rate and lower death rate. After the comparison of Lychnis fulgens diploid and tetraploid plant guard cell chloroplast numbers, the test results agree with the chromosome counting detection. That confirmed guard cell chloroplast counting were an effective method of determine level of Lychnis fulgens ploidy level. |
